抑郁症模型小鼠肠道菌群的微生物生态学研究 被引量:1

摘要 目的:基于16S rDNA宏基因分类测序分析,通过检测抑郁症造模成功小鼠的粪便标本,探讨抑郁症小鼠肠道菌群的微生物生态学特点。方法:选择4周龄SPF级雌性小鼠,并随机分组;通过皮下注射苯甲酸雌二醇造模;记录实验组与对照组小鼠实验过程体重、饮食、学习记忆能力变化并进行统计分析,确定造模效果;采集造模成功小鼠与正常对照小鼠粪便标本,进行粪便菌群宏基因组检测与分析。结果:小鼠造模后呈现明显的抑郁样表现,主要表现在体重增加速度减慢、饮食欲望减退、学习记忆能力明显下降以及菌群多样性降低,g_unclassified_Enterobacteriaceae和g_Roseburia两个属在实验组与对照组间差异显著(P<0.05)。结论:小鼠长期处于高雌激素的病理状态会导致抑郁,并且抑郁症小鼠相较正常小鼠存在肠道菌群紊乱现象。 Objective:Based on 16S rDNAacer gene classification and sequencing analysis,the fecal samples of mice with depression were detected to explore the microbial ecology characteristics of intestinal microflora of mice with depression.Methods:Female mice of SPF grade at 4 weeks of age were selected and randomly assigned.The model was made by subcutaneous injection of estradiol benzoate;The experimental results of body weight,diet,learning and memory ability of experimental group and control group were recorded,and statistical analysis was made to determine the modeling effect;Fecal specimens of mice with successful modeling and normal control were collected and sent for metagenomic detection and analysis of fecal flora.Results:The mice show obvious depression-like performance after modeling,mainly manifested in slower weight gain,decreased appetite for food,significantly decreased learning and memory ability,and decreased diversity of bacteria.Compared with normal mice,g_Unclassified_Enterobacteriaceae and g_Roseburia were significantly different between the experimental group and the control group(P<0.05).Conclusion:Long-term exposure to high estrogen levels in mice leads to depression,and the intestinal flora of depressed mice is disturbed compared with normal mice.
