转基因聚球藻7942的vp19基因表达效率及其光合生理特性研究 被引量:1

摘要 采用荧光定量PCR方法检测转基因聚球藻7942在不同生长阶段vp19基因的相对表达量,在此基础上,通过氧电极的方法测定转vp19基因聚球藻7942在不同温度、光强、pH和盐度下光合活性的变化,探究其最适生长条件。结果表明:转基因聚球藻7942的最佳采收时间是对数生长后期(约12 d),其vp19基因相对表达率达到9.2%,VP19蛋白的产率达到7.31 mg∕L。转vp19基因型聚球藻的最适生长条件为:温度38℃,光照强度2500 lx,pH 8.0,盐度0 mol∕L NaCl。研究结果可为转vp19基因型聚球藻口服疫苗规模制备提供依据。 Fluorescent quantitative PCR was used to detect the relative expression of vp19 gene in different growth stages of transgenic Synechococcus sp.PCC 7942.On this basis,the changes of photosynthetic activity of transgenic Synechococcus sp.PCC 7942 under different temperature,light intensity,pH and salinity were determined by oxygen electrode method to explore the optimal growth conditions.The results showed that the best harvest time of transgenic Synechococcus sp.PCC 7942 was the late logarithmic growth period(about 12 days),the relative expression rate of vp19 gene was 9.2%,and the yield of VP19 protein was 7.31 mg/L.The optimum growth conditions for transgenic Synechococcus sp.PCC 7942 were as follows:temperature 38℃,light intensity2500 lx,pH 8.0,salinity 0 mol/L NaCl.The results could provide a basis for the scale preparation of oral vaccine with transgenic vp19 Synechococcus sp.PCC 7942.
