摘要 目的研究长链非编码RNA(LncRNA)H19对雨蛙素刺激的大鼠胰腺腺泡细胞AR42J细胞凋亡的影响。方法在体外通过用雨蛙素刺激AR42J细胞建立AP模型,采用qRT-PCR和Western blot检测沉默以及过表达H19后凋亡相关基因Caspase 3及Caspase 9的mRNA和蛋白表达水平;流式细胞术检测各组细胞的凋亡率。结果H19沉默后,凋亡相关基因Caspase 3及Caspase 9的mRNA和蛋白水平下降,细胞凋亡率也相对减少(P<0.05);而过表达H19后,Caspase 3及Caspase 9的mRNA和蛋白水平增加,细胞凋亡率也是升高的(P<0.05)。结论沉默LncRNA H19可减少雨蛙素诱导的胰腺腺泡细胞凋亡,提示其可能成为治疗急性胰腺炎的潜在有效靶点。 Objective To investigate the effect of long non-coding RNA(LncRNA)H19 on the apoptosis of rat pancreatic acinar cells AR42J induced by caerulein.Methods An AP model in vitro was established by stimulating with caerulein.The mRNA and protein expression levels of Caspase 3 and Caspase 9 were detected by qRT-PCR and Western blot after H19 silencing and overexpression.The apoptosis rate of AR42J cells in each group was detected by Annexin V-FITC/PI assay.Results After H19 silencing,the mRNA and protein levels of Caspase 3 and Caspase 9 significantly decreased,and the apoptosis rate was also relatively reduced(P<0.05).After H19 overexpression,the mRNA and protein levels of Caspase 3 and Caspase 9 significantly increased,and the apoptosis rate also significantly increased(P<0.05).Conclusion The silencing of LncRNA H19 can significantly reduce the apoptosis of cerulean-induced pancreatic acinar cells,suggesting that it may be a potential effective target for the treatment of acute pancreatitis.
