不同贮藏温度和时间对思茅松花粉活力的影响 被引量:3

Effects of Different Storage Temperatures and Time on Pollen Viability of Pinus kesiya var.langbianensis

摘要 为探讨思茅松(Pinus kesiya var.langbianensis)花粉的贮藏特性,以思茅松种子园无性系花粉为材料,采用离体萌发法研究4种贮藏方式对花粉活力的影响。结果表明,以100 mg/L H3BO3溶液为培养基,在28℃恒温条件下暗培养,12 h可观测到花粉萌发,48 h花粉已具有较好的萌发识别度,48、60和72 h萌发率差异不显著,48 h是该试验条件下花粉活力测定的最佳时间;4种贮藏方式0.5年时,花粉活力比保存前降低29.8%~69.7%,1年时花粉活力降低59.8%~96.4%;低温干燥比常温干燥更有利于保持花粉活力,3种低温干燥保存方式间花粉活力差异不显著。低温干燥保存1年的思茅松花粉活力能满足生产的要求。 In order to explore storage characteristics of Pinus kesiya var.langbianensis pollens,taking clone pol⁃lens from P.kesiya var.langbianensis seed ochards as materials,effects of four storage methods on pollen viabil⁃ity were studied by in vito method.Results showed that pollen germination was observed at 12 h after dark cul⁃ture at 28℃with 100 mg/L H3BO3 solution as substrate.Pollen germination recognition was good at 48 h.There was no significant difference in germination rate among 48,60 and 72 h.48 h was the best time for pollen viabil⁃ity determination under this experimental condition.Pollen viability decreased by 29.8%-69.7%at half year and 59.8%-96.4%at one year.Pollen viability could be sustained in low temperature and dry condition better than that of room temperature and dry condition.There was no significant difference in pollen viability among three storage methods with low temperature and dry condition.Pollen viability of P.kesiya var.langbianensis stored at low temperature and dry condition for 1 year could meet requirements of production.
