缺氧诱导因子1在下咽癌中的表达及对细胞生物学功能的影响

摘要 目的检测缺氧诱导因子1(hypoxia inducible factor-1,HIF-1)在下咽癌中的表达及对下咽癌细胞生物学功能的影响。方法应用real-time PCR技术检测30例下咽癌组织和对应的癌旁组织中HIF-1的表达水平;将HIF-1过表达载体转染至下咽癌细胞FaDu细胞中,并通过观察绿色荧光蛋白、Western blot和real-time PCR技术检测转染效率;Transwell小室实验检测细胞迁移能力;CCK8法检测细胞增殖。结果 HIF-1在下咽癌组织中高表达,表达量为1.436±0.2575,在癌旁组织中的表达量为0.7337±0.2575,差异有统计学意义(t=2.725,P<0.05),以上结果与TCGA数据库分析结果一致。转染HIF-1过表达组发生转移的细胞数量(78.33±10.84)较对照组(35±10.84)增多,差异有统计学意义(t=3.999,P<0.05);转染HIF-1过表达组细胞增殖能力(OD_(72h)=1.64,OD_(96h)=1.85)均高于对照组(OD_(72h)=1.34,OD_(96h)=1.59),差异有统计学意义(t=1.715,P<0.05)。结论 HIF-1促进下咽癌细胞FaDu的迁移和增殖能力,并参与下咽癌的转移。 OBJECTIVE To detect the expression of HIF-1 in hypopharyngeal cancer and its effect on the biological function of hypopharyngeal cancer cells.METHODS Real-time PCR was used to detect the expression of HIF-1 in 30 pairs of hypopharyngeal cancer tissues.The HIF-1 overexpression vector was transfected into hypopharyngeal cancer cell FaDu cells,and the transfection efficiency was detected by Western blot and real-time PCR.CCK8 assay was used to detect cell proliferation;Transwell chamber assay to detect cell migration ability.RESULTS The expression of HIF-1 in hypopharyngeal carcinoma was 1.436±0.2575,and the expression in normal tissues adjacent to cancer was 0.7337±0.2575.HIF-1 was highly expressed in hypopharyngeal carcinoma,and the difference was statistically significant(P=0.0086,P<0.05);these results are consistent with the results of the TCGA database analysis.The number of metastatic cells in the transfected HIF-1 overexpression group was 78.33±10.84,which was more than the control group(35±10.84),the difference was statistically significant(P<0.05);the cell proliferation ability of the transfected HIF-1 overexpression group(OD_(72h)=1.64,OD_(96h)=1.85) was higher than the control group(OD_(72h)=1.34,OD_(96h)=1.59),and the difference was statistically significant (P<0.05).CONCLUSION HIF-1 promoted the migration and proliferation ability of hypopharyngeal cancer cell,which is closely related to the metastasis of hypopharyngeal cancer.
