柴油废气颗粒体外对鼻息肉组织单个细胞炎性因子释放的影响

Effect of diesel exhaust particle on the inflammatory cytokine production in dispersed nasal polyp cells in vitro

摘要 目的观察柴油废气颗粒(diesel exhaust particle,DEP)体外对慢性鼻窦炎合并鼻息肉(chronic rhinosinusitis with nasal polyps,CRSwNP)患者鼻息肉(nasal polyps,NP)组织炎性因子生成的影响。方法收集CRSwNP患者NP组织,制备NP组织单个细胞,体外分别用25、50和100μg/ml DEP刺激,24 h后检测细胞活性和细胞培养上清中Th1细胞(IL-12p70、IFN-γ),Th2细胞(IL-4、IL-5、IL-13)和Th17细胞(IL-17、IL-1β、TNF-α)的浓度。结果与未刺激组相比,50μg/ml和100μg/ml DEP刺激组中IFN-γ、IL-4、IL-13、IL-1β和TNF-α浓度均显著降低,100μg/ml DEP刺激组中IL-5和IL-17显著降低。活细胞百分比和IL-12p70的浓度在各组间没有显著差异。结论DEP能抑制NP组织单个细胞多种炎性因子的生成,而且这种抑制作用与细胞毒性无关。 OBJECTIVE To investigate the impact of diesel exhaust particle(DEP)on the production of inflammatory cytokines in nasal polyp(NP)tissue.METHODS The dispersed NP cells from patients with chronic rhinosinusitis with nasal polyps(CRSwNP)were prepared and stimulated with 25,50 and 100μg/ml DEP respectively.Cell viability and the concentrations of Th1(IL-12p70,IFN-γ),Th1(IL-4,IL-5,IL-13)and Th17(IL-17,IL-1β,TNF-α)cytokines in the supernatants of cell culture were measured 24 hours after stimulation.RESULTS Compared to the un-stimulated group,the concentrations of IFN-γ,IL-4,IL-13,IL-1βand TNF-αwere significantly lower in dispersed NP cells stimulated by 50μg/ml and 100μg/ml DEP;and the concentrations of IL-5 and IL-17 were significantly lower in cells stimulated by 100μg/ml DEP.However,the percentage of cell viability and the IL-12p70 concentration were comparable between groups.CONCLUSION DEP inhibits the production of most inflammatory cytokines in NP tissue in vitro and this inhibition was not caused by cell toxicity.
