摘要
目的探讨脂肪间充质干细胞(ADSCs)移植对放射性碘131(^(131)I)诱导后大鼠甲状腺功能减退的影响。方法将40只6周龄雄性SD大鼠随机分为4组:阴性对照组、^(131)I模型组、药物对照组和ADSCs治疗组,每组10只。采用酶联免疫吸附法检测各组大鼠促甲状腺激素(TSH)、游离三碘甲状腺原氨酸(FT3)、游离甲状腺素(FT4)、肿瘤坏死因子-α(TNF-α)、白细胞介素6(IL-6)及白细胞介素10(IL-10)水平。应用甲状腺^(131)I静态显像评估甲状腺摄碘功能。结果^(131)I模型组大鼠的TSH、TNF-α、IL-6水平均高于阴性对照组,FT3、FT4、IL-10水平均低于阴性对照组,差异均有统计学意义(P<0.05)。ADSCs治疗组大鼠的TSH、TNF-α、IL-6水平均低于^(131)I模型组,差异均有统计学意义(P<0.05);ADSCs治疗组大鼠的FT3、IL-10水平均高于^(131)I模型组,但差异无统计学意义(P>0.05)。药物对照组与ADSCs治疗组大鼠的TSH水平均低于^(131)I模型组,差异有统计学意义(P<0.05)。甲状腺^(131)I静态显像结果显示,ADSCs治疗组轻微可见甲状腺显影,而药物对照组与^(131)I模型组甲状腺组织均未见显影。结论ADSCs移植可能会促进甲状腺激素的分泌,改善甲状腺的摄碘功能,对于放射性^(131)I诱导后大鼠甲状腺功能减退具有一定的潜在治疗作用。
Objective To investigate the effect of adipose derived mesenchymal stem cells(ADSCs)transplantation on hypothyroidism induced by ^(131)I in rats.Methods Forty 6-week-old male SD rats were randomly divided into four groups:negative control group,^(131)I model group,drug control group and ADSCs treatment group,with 10 rats in each group.The levels of thyroid stimulating hormone(TSH),free triiodothyronine(FT3),free thyroxine(FT4),tumor necrosis factor(TNF-α),interleukin-6(IL-6)and interleukin-10(IL-10)were detected by enzyme linked immunosorbent assay.Thyroid iodine uptake was evaluated by ^(131)I static imaging.Results TSH and TNF in ^(131)I model group、The level of IL-6 was higher than that of the negative control group,and the levels of FT3,FT4 and IL-10 were lower than those of the negative control group(P<0.05).TSH and TNF levels in ADSCs treated rats、The level of IL-6 in model group was significantly lower than that in ^(131)I group(P<0.05);The levels of FT3 and IL-10 in ADSCs treatment group were higher than those in ^(131)I model group,but the difference was not statistically significant(P>0.05).The TSH levels of the drug control group and ADSCs group were lower than that of the ^(131)I model group,the difference was statistically significant(P<0.05).The results of thyroid ^(131)I static imaging showed that there was slight thyroid development in ADSCs treatment group,but no thyroid development in drug control group and ^(131)I model group.Conclusion ADSCs transplantation may increase the secretion of thyroid hormone and improve the iodine uptake function of thyroid.It has a potential therapeutic effect on the hypothyroidism induced by ^(131)I in rats.
