响应曲面法优化白及挥发性成分提取工艺 被引量:3

摘要 以提取率作为响应值,在单因素试验的基础上采用响应曲面法研究提取粉碎粒度、压力、温度三个因素对白及挥发油提取效果的影响,确定挥发油最佳提取工艺;运用气相色谱-质谱联用仪(GC-MS)对其主要化学成分进行鉴定。结果表明:粉碎粒度、提取压力、提取温度分别为50目、27 MPa、50℃时,提取率达到最大(1.28%);通过NIST14质谱库检索,鉴定白及挥发油中匹配度大于85的72种可能物质,运用峰面积归一法确定各个组分的相对含量,鉴定物质占挥发油总量的92%。 Based on single factor testing,the response surface method was used to study the influence of the crushing particle size,pressure and temperature on the extraction of volatile oil from Bletilla striata in order to identify the best process for extraction of such oil.Gas chromatography-mass spectrometry(GC-MS)was then used to analyze the chemical composition of the oil extracted.The best extraction conditions for maximising the extraction rate proved to be:particle size 50 mesh,extraction pressure of 27 MPa and extraction temperature of 50℃.In the volatile oil extracted,72 compounds were identified by a NIST14 mass spectral library search,with the content of each component determined by the peak area unification method.Together these identified compounds accounted for 92%of the total volume of the volatile oil.
