景天三七总多酚提取工艺的优化及活性研究 被引量:12

摘要 采用超声提取景天三七中的多酚,并利用Box-Behnken响应面法对影响景天三七总多酚提取率的因素(液料比、乙醇浓度、超声时间、超声温度)进行了优化,并以DPPH自由基和ABTS自由基清除能力为评价指标,研究景天三七总多酚体外抗氧化活性。结果显示,最佳的提取条件为乙醇浓度63%,液料比36∶1(mL·g^(-1)),超声温度65℃,超声时间27 min。此条件下多酚的提取率为(28.08±0.21)mg·g^(-1)。此条件下得到的景天三七总多酚对DPPH自由基和ABTS自由基具有不同的清除能力,其半数抑制浓度(IC50)分别为0.81μg·mL^(-1),9.02μg·mL^(-1)。本研究为景天三七的资源开发提供理论基础。 The total polyphenols from Sedum aizoon L.were extracted by ultrasonic extraction.The effects of extraction conditions(ethanol concentration,liquid/solid ratio,ultrasonic time,ultrasonic temperature)on the extraction rate of total polyphenol from Sedum aizoon L.were optimizedby Box-Behnken response surface methodology.Meanwhile,the antioxidant activities of total polyphenols from Sedum aizoon L.in vitro were analyzedwiththe indexes of DPPH and ABTS free radical scavenging ability.The results showed that the optimal extraction conditions were determined as follows:ethanol concentration of 63%,liquid/solid ratio of 36∶1(mL·g^(-1)),ultrasonic time of27 min,ultrasonic temperature of 65℃.Under the optimal extraction conditions,the content of total polyphenols was(28.08±0.21)mg·g^(-1).Total polyphenols from Sedum aizoon L.presented different DPPH and ABTS scavenging activities,and the half inhibitory concentration(IC 50)was 0.81μg·mL^(-1),9.02μg·mL^(-1),respectively.This study could provide a theoretical basis for resource development of Sedum aizoon L.
