浙江与广西产三叶青水煎液体内肺癌抑制作用的对比研究

Comparison of Inhibitory Effects of Sanyeqing(Tetrastigma Hemsleyanum Diels et Gilg)from Zhejiang and Guangxi Water Extra on Lung Cancer

目的:比较浙江产三叶青与广西产三叶青肺癌抑制效果。方法:以三叶青水煎液处理A549肺癌荷瘤裸鼠,测定肿瘤体积和重量,计算抑瘤率,采用TUNEL法测定细胞凋亡诱导效果,采用免疫组化法测定凋亡相关蛋白表达。结果:浙江产三叶青水煎液能显著抑制肿瘤体积与质量的增长(P<0.05或P<0.01),抑瘤效果显著,且能够显著诱导肿瘤细胞凋亡;广西产三叶青水煎液肿瘤抑制、肿瘤细胞凋亡诱导效果不显著(P>0.05);且浙江产三叶青水煎液能显著调节凋亡相关蛋白的表达,上调BAX表达,下调Bcl-2表达,增强Caspase3及Caspase9表达增强(P<0.05或P<0.01);广西产三叶青蛋白表达调节效果不显著(P>0.05)。结论:浙江产三叶青体内肺癌抑制效果显著优于广西产三叶青,为三叶青临床应用及标准制定提供一定的数据支持。

Objective:To compare the difference of inhibitory effects of T.hemsleyanum from Zhejiang and Guangxi water extra on lung cancer.Methods:A549 lung cancer bearing nude mice were given by T.hemsleyanum water extra for 1 week,the volume,weight of tumor tissue were measured,and inhibition rate was calculated,the induction effect of apoptosis was determined by TUNEL method,and the expression of apoptosis related proteins were determined by immunohistochemistry.Results:T.hemsleyanum water extract from Zhejiang Province could significantly inhibit the growth of tumor and induce apoptosis of tumor cells(P<0.05 or P<0.01),the effects of T.hemsleyanum water extract from Guangxi were no significant(P>0.05).T.hemsleyanum from Zhejiang could significantly regulate the expressions of apoptosis related proteins,up-regulate Bax and down-regulate Bcl-2,enhance the expressions of Caspase-3 and Caspase-9(P<0.05 or P<0.01),the regulation effects of T.hemsleyanum from Guangxi on above proteins were no significant(P>0.05).Conclusion:The inhibition effect of T.hemsleyanum from Zhejiang on ling cancer were more significant than that of T.hemsleyanum from Guangxi,which will provide important data support for clinical application and standard formulation.
