奶牛黏膜病的BVDV HJ-1株全基因组序列分析 被引量:2

摘要 BVDV HJ-1分离株属于1b基因亚型是引起奶牛发生黏膜病和出血综合症病原,导致高病死率。为了解BVDV HJ-1株的遗传学特征,研究应用PCR方法分13段扩增其全基因组序列,通过克隆测序最终组装出全序列。应用生物信息学软件对BVDV HJ-1基因序列同源性、跨膜结构域、糖基化位点、5’UTR一级结构与二级结构的差异等进行分析。结果测得该毒株全序列为12282 nt,与参考毒株相比存在高度变异,序列同源性在70.1%~93.5%,尤其是非结构蛋白NS4A、NS4B和NS5A同源性在49.0%~93.0%。E1-E2存在5个潜在的跨膜信号区,结构蛋白上富含14个潜在的糖基化位点,5’UTR Ⅰ区和Ⅲb-Ⅲe区相对保守,不同毒株之间II区的二级结构变异大。总之,BVDV HJ-1株全基因组序列信息丰富了我国BVDV的遗传学数据资源,为下一步疫苗的研制和致病机理的研究奠定了基础。 BVDV HJ-1 isolate belonging to gene subtype 1b,which caused mucosal disease and bleeding syndrome in dairy cows,resulting in high mortality.To understand the genetic characteristics of BVDV HJ-1 strain,the whole genome sequence was amplified in 13 segments by PCR method,and the complete sequence was finally assembled with soft by sequencing results.The bioinformatics analysis software was used to analyze the differences between BVDV HJ-1 gene sequence homology,transmembrane domain,glycosylation sites,5’UTR primary and secondary structures.The results showed that the total sequence of the strain was 12282 nt,and the sequence homology was 70.1%-93.5%,the homology of nonstructural protein NS4A,NS4B and NS5A was 49.0%-93.0% compared with the reference strain.E1-E2 had five potential transmembrane signal regions,and the potential glycosylation sites had 14 from structural proteins.The UTR Ⅰ and Ⅲb-Ⅲe regions were relatively conserved,and the secondary structure variation of the II regions between different strains was large.To sum up,the whole genome sequence information of BVDV HJ-1 strain enriched the genetic data resources of BVDV in China and laid a foundation for the development of vaccine and the study of pathogenic mechanism.
