灯盏细辛多酚纯化工艺的响应面法优化及其抑菌活性研究 被引量:2

Purification Process Optimization of Polyphenols from Erigeron breviscapus by Response Surface Analysis and Their Antibacterial Activities

摘要 利用单因素与响应面试验确定大孔树脂纯化灯盏细辛多酚提取物的工艺。通过静态吸附-洗脱试验,比较6种不同类型大孔树脂对目标化合物的吸附、解吸性能后,利用动态吸附-洗脱试验确定最佳纯化工艺,同时采用滤纸片法考察纯化后的多酚化合物对大肠杆菌、枯草杆菌和金黄色葡萄球菌的抑菌活性。结果表明,H103型树脂对灯盏细辛多酚提取物的纯化工艺条件为:上样液质量浓度为1.9 mg/mL,pH为5.1,上样液体积为80 mL,上样流速为2 mL/min,洗脱剂乙醇体积分数为69%,体积为120 mL,洗脱流速为1 mL/min,产物的多酚纯度由11.25%提高至54.76%。纯化后的灯盏细辛多酚对3种受试菌具有不同程度的抑制活性,其中对金黄色葡萄球菌的抑制活性最强,因此可为灯盏细辛多酚的后续开发利用提供参考。 The purified technologies of polyphenols extracted from Erigeron breviscapus through macroporous resins were investigated by single factor tests and response surface analysis.The adsorption and desorption performances of six macroporous resins types were compared by static adsorption-desorption experiments,then the optimal purification process was determined by dynamic adsorption-desorption experiments.Meanwhile,their bacteriostatic activities against Escherichia coli,Staphylococcus aureus,and Bacillus subtilis were detected by filtering paper method.Results showed that the purification conditions of polyphenols through H103 resin were determined as follows:sample solution concentration of 1.9 mg/mL,pH value of 5.1,the sample volume of 80 mL,loading speed of 2 mL/min,ethanol volumefrac of 69%,eluent volume of 120 mL,and elution velocity of 1 mL/min.Under the conditions,the purity of product was increased from 11.25%to 54.76%.Besides,the purified polyphenols of Erigeron breviscapus showed inhibitory effects with varying degrees on the three tested bacteria and presented the strongest inhibitory activity against Staphylococcus aureus.Therefore,these data could provide reference for the further exploitation and utilization of polyphenols of Erigeron breviscapus.
