摘要
目的挖掘卵巢子宫内膜异位症病灶组织差异基因,探索黏附相关基因在卵巢内异症发生发展过程中的作用。方法利用美国国立生物技术信息中心的Gene Expression Omnibus数据库,检索卵巢内异症表达谱芯片。通过在线GEO2R工具找到差异基因,并利用功能注释生物信息学芯片分析技术找到黏附相关基因。结果最终纳入1个卵巢内异症相关基因芯片,找到差异表达基因共568个,其中THBS1、VWF、THBS2、COMP、COL5A1、COL6A2、ITGA7和ITGB8这8个基因与黏附功能密切相关,并主要在细胞外基质受体相互作用、局灶性粘连和PI3K-Akt这3条通路中起重要作用。结论卵巢内异症病灶中黏附相关基因表达存在异常,介导相关信号通路异常活化,从而促进卵巢内异症的发生发展,为经血逆流学说提供了基因角度证据。
Objective To explore the differential genes of ovarian endometriosis lesions and explore the role of adhesion-related genes in the occurrence and development of ovarian endometriosis.MethodsGene Expression Omnibus(GEO)database of National Biotechnology Information Center(NCBI)was used to retrieve the expression profile chip of ovarian endometriosis.Differential genes were identified by online GEO2R analysis,and adhesion-related genes were identified by Functional Annotation Bioinformatics Microarray Analysis(DAVID).GEO2R was used to analyze the expression of each adhesion-related gene,and related pathways were mined through online KEGG database.ResultsA total of 568 differentially expressed genes were found,including THBS1,VWF,THBS2,COMP,COL5A1,COL6A2,ITGA7 and ITGB8 closely related to adhesion function.These adhesion genes were found to play an important role in ECM-receptor interaction,Focal adhesions and PI3K-Akt signaling pathway through online KEGG database.ConclusionAbnormal expression of adhesion-related genes in ovarian endometriosis lesions mediates abnormal activation of related signaling pathways,thereby promoting the development of ovarian endometriosis and providing genetic evidence for the study of Retrograde Menstruation.
