长链非编码RNA LINC00467靶向miR-1247-5p调控肝癌细胞增殖、迁移及侵袭的影响 被引量:3

摘要 目的探究长链非编码RNA LINC00467(lncRNA LINC00467)靶向微小RNA-1247-5p(miR-1247-5p)对肝癌细胞增殖、迁移及侵袭的影响及其作用机制。方法体外培养肝癌细胞株HepG2、SMMC-7721、BEL-7402与正常肝细胞株L02,实时荧光定量PCR(qRT-PCR)检测细胞中lncRNA LINC00467与miR-1247-5p相对表达量。用LINC00467 siRNA(si-LINC00467组)、miR-1247-5p mimic(miR-1247-5p组)及其各自阴性对照分别转染肝癌HepG2细胞,噻唑蓝(MTT)检测细胞增殖活性,Transwell实验检测HepG2细胞迁移及侵袭能力,荧光素酶报告基因实验确定LINC00467与miR-1247-5p的靶向关系,Western印迹检测细胞增殖、迁移及侵袭相关蛋白表达。结果与正常肝细胞株L02相比,不同肝癌细胞株HepG2、SMMC-7721、BEL-7402中LINC00467相对表达量明显升高(P<0.05),而miR-1247-5p相对表达量明显降低(P<0.05);沉默LINC00467或miR-1247-5p过表达均可显著抑制肝癌细胞增殖及细胞周期蛋白(Cyclin)D1的表达,显著促进p21及p27蛋白表达(P<0.05);沉默LINC00467或miR-1247-5p过表达还能显著抑制肝癌细胞迁移、侵袭及基质金属蛋白酶(MMP)-2、MMP-9、MMP-14的表达(P<0.05);LINC00467可负向调节miR-1247-5p表达;抑制miR-1247-5p可显著减弱si-LINC00467对肝癌细胞增殖、迁移及侵袭能力的作用(P<0.05)。结论lncRNA LINC00467可通过靶向miR-1247-5p增强肝癌细胞增殖、迁移及侵袭能力。
