CHD1L对胃癌细胞增殖、侵袭及转移的影响 被引量:2

摘要 目的研究胃癌细胞生物学行为是否受沉默CHD1L的调控。方法(1)筛选沉默效率最高的干扰组进行后续试验;(2)蛋白p53、p21、Nur77、ARHGEF9在沉默CHD1L后表达水平利用Western Blot免疫印迹进行测定;(3)CHD1L沉默对胃癌细胞凋亡和增殖的影响分别采用流式细胞术和MTT比色法测定;(4)沉默CHD1L后对胃癌上皮细胞-间充质转化的影响采用Transwell小室侵袭、细胞划痕愈合实验测定。结果(1)CHD1L-shRNA-1沉默效果最好,表达量下调58.60%;(2)p53、p21、Nur77、ARHGEF9表达量伴随CHD1L的沉默呈现明显上升趋势;(3)胃癌MGC-823细胞中CHD1L被沉默后S、G1期细胞数量呈现明显的减少与增多趋势,细胞增殖的抑制率和凋亡数量与时间呈正相关(P<0.05);(4)胃癌MGC-823中CHD1L被沉默后穿膜细胞数量明显减少为(58.63±10.97)个、细胞迁移距离缩短为(0.54±0.34)μm(P<0.05)。结论MGC-823细胞的侵袭力和迁移能力在沉默CHD1L受到明显的抑制。 Objective To study whether the biological behavior of gastric cancer cells is regulated by silencing CHD1L.Methods(1)Screen the interference group with the highest silencing efficiency for follow-up experiments;(2)Expression levels of related proteins p53,p21,Nur77,and ARHGEF9 that inhibit the growth and development of gastric cancer after silencing CHD1L Western blotting was used for the determination;(3)The effects of CHD1L silencing on the apoptosis and proliferation of gastric cancer cells were determined by flow cytometry and MTT colorimetry;(4)The effect of silencing CHD1L on gastric cancer epithelial cell-mesenchymal transition was determined by the Transwell chamber invasion and cell scratch healing test.Results(1)CHD1L-shRNA-1 has the best silencing effect,and the expression of CHD1L in gastric cancer cells is down-regulated by 58.60%;(2)The expression of p53,p21,Nur77,and ARHGEF9 protein shows a significant upward trend with the silencing of CHD1L;(3)Compared with the control group,after CHD1L was silenced in gastric cancer MGC-823 cells,the number of S and G1 phase cells showed a significant decrease and increase trend.The inhibition rate of cell proliferation and the number of apoptosis were positively correlated with time(P<0.05);(4)Compared with the control group,the number of transmembrane cells in gastric cancer MGC-823 after CHD1L was silenced was significantly reduced to 58.63±10.97,and the cell migration distance was shortened to(0.54±0.34)μm(P<0.05).Conclusion The invasiveness and migration ability of MGC-823 cells was significantly inhibited by silencing CHD1L.
