薏苡仁提取物对人皮肤原代成纤维细胞MMP1/3 mRNA表达的影响 被引量:2

Effects of Extraction of Semen Coicis on the Expression of MMP1,MMP3 mRNA Induced by Ultraviolet B Radiation in Human Dermal Fibroblasts

摘要 目的探讨薏苡仁提取物(ESC)对中波紫外线(UVB)照射、体外培养人皮肤成纤维细胞(HDF)的基质金属蛋白酶1、3(MMP1、MMP3)mRNA表达的影响。方法采用酶消化法和组织块法培养HDF。四甲基偶氮唑盐微量酶反应比色法(MTT)检测ESC对HDF增殖的影响。用逆转录多聚酶链反应(RT-PCR)检测1、2.5、5μL/mL ESC及基质对中波紫外线(UVB)照射后HDF中MMP1、MMP3 mRNA表达水平的变化。结果UVB照射可以显著降低HDF中MMP1、MMP3 mRNA的表达,差异有统计学意义(P<0.05)。30 mJ/cm^(2) UVB照射细胞组加入不同浓度基质及ESC,MMP1、MMP3 mRNA与对照组的差异有统计学意义(P<0.05)。结论ESC可有效降低UVB照射后的HDF中MMP1、MMP3 mRNA表达,且有剂量-效应关系。为后续研究抗皮肤光老化提供基础。 Objective In order to investigate the expression of matrix metallo proteinase 1,3(MMP1,MMP3)of human dermal fibroblasts(HDF)induced by ultraviolet B(UVB)radiation,and to explore the protective effect of extraction of semen coicis(ESC)on the expression of MMP1,3 of HDF induced by UVB radiation.Methods Colorimetric experiment of diphenyltetraazawa bromide was performed to detected the effects of ESC on the proliferation of HDF.Then,the effects of ESC treatment on mRNA of MMP1,MMP3 in UVB radiated fibroblasts were assessed.Results ESC exhibited no inhibitory effect on the proliferation of fibroblasts.The expression of MMP1,MMP3 mRNA in different dose of UVB radiated fibroblasts were significant different(P<0.05),especially in 30 mJ/cm2 UVB group.ESC treatment exhibited down-regulation on the expression ofMMP1,MMP3mRNA in 30mJ/cm2UVB radiated fibroblasts(P<0.05).Conclusion ESC have no influence on the proliferation of fibroblasts and can effectively down-regulate the expression of MMP1,MMP3 mRNA of HDF.ESC could remit photo damaging induced by UVB.
