赤芍多糖的提取纯化工艺与抗氧化活性研究 被引量:3

摘要 建立赤芍多糖的提取方法,并对其进行分离和纯化。选择超声提取的方法,首先对赤芍中多糖分别采用热水和碱液作为溶剂进行提取,对得到的水提液和碱提液采用醇沉法进行多糖的初步分离和纯化,得到两个部位的粗多糖P80和PB,提取率分别为10.92%和3.69%,用苯酚-硫酸法测得P80和PB的糖含量分别为52.49%和34.49%。选择糖含量较高的P80进行Sevag法除去蛋白及透析等操作,冻干得到PRP80,随后使用DEAE-Cellulose 52阴离子交换柱层析对其进行纯化,得到PRP80-1和PRP80-2。通过测定P80和PB对2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ATBS)自由基和1,1-二苯基-2-三硝基苯肼(DPPH)自由基的清除率来考察赤芍粗多糖的体外抗氧化活性,结果表明P80和PB均具有良好的抗氧化活性。 To establish the extraction and purification method of polysaccharides from Radix Paeoniae Rubra. Firstly, hot water and alkaline solution were used as solvent to extract polysaccharides from Radix Paeoniae Rubra. Then, the water extract and alkaline extract were separated and purified by alcohol precipitation method. The crude polysaccharides P80 and PB were obtained. The extraction rate was 10.92 % and 3.69 %, respectively. The sugar content of P80 and PB was52.49 % and 34.49 %, respectively by phenol sulfuric acid method. P80 with high sugar content was selected for protein removal by sevag method and dialysis.PRP80 was obtained by freeze-drying. Then PRP8080-1 and PRP8080-2 were obtained by DEAE cellulose 52 anion exchange column chromatography. The antioxidant activities of crude polysaccharides from Radix Paeoniae Rubra were studied by measuring the scavenging rates of P80 and PB on 2,2-diazo-bis(3-ethyl-benzothiazol-6-sulfonic acid) diammonium salt(ATBS) and 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH) radicals. The results showed that P80 and PB had good antioxidant activities.
