白藜芦醇通过抑制SIRT3/β-catenin/PPARγ信号通路对急性心肌梗死大鼠心肌细胞凋亡的影响

摘要 目的:探究白藜芦醇通过抑制沉默信息调节因子3(Silent Information Regulator 3,SIRT3)/β-连环蛋白(β-catenin)/过氧化物酶体增殖物激活受体γ(Peroxisome Proliferator Activated Receptorγ,PPARγ)信号通路对急性心肌梗死(Acute Myocardial Infarction,AMI)大鼠心肌细胞凋亡。方法:45只SD大鼠随机分为3组(n=15):假手术组,AMI组和白藜芦醇组。AMI组和白藜芦醇组建立AMI模型。白藜芦醇组大鼠舌静脉注射白藜芦醇干预。检测和比较各组大鼠心肌细胞损伤、凋亡情况并比较SIRT3、β-catenin、PPARγ蛋白水平。结果:AMI组大鼠细胞形状发生改变,排列紊乱,并出现炎性浸润和细胞坏死。白藜芦醇组细胞形态和排列恢复,细胞坏死显著减少。AMI组大鼠血清cTnT、CK-MB、IL-6和TNF-α水平显著高于假手术组(P<0.05),白藜芦醇组大鼠血清cTnT、CK-MB、IL-6和TNF-α水平显著低于AMI组(P<0.05)。AMI组大鼠心肌细胞凋亡率显著高于假手术组(P<0.05),白藜芦醇组大鼠心肌细胞凋亡率显著低于AMI组(P<0.05)。AMI组大鼠心肌组织中SIRT3、β-catenin、PPARγ蛋白水平高于假手术组(P<0.05),白藜芦醇组大鼠心肌组织中SIRT3、β-catenin、PPARγ蛋白水平显著低于AMI组(P<0.05)。结论:白藜芦醇通过抑制SIRT3/β-catenin/PPARγ通路缓解AMI引起的心肌细胞凋亡,并减少炎症反应改善心肌功能。 Objective:To investigate resveratrol by inhibiting silent information regulator 3(SIRT3)/β-catenin/peroxisome proliferator activated receptorγ(PPARγ)signaling pathway in cardiomyocyte apoptosis in acute myocardial infarction(AMI)rats.Methods:Forty-five SD rats were randomly divided into 3 groups(n=15):sham operation group,AMI group and resveratrol group.An AMI model was established in the AMI group and the resveratrol group.Rats in the resveratrol group were treated with intravenous injection of resveratrol.The injury and apoptosis of cardiomyocytes in each group were detected and compared,and the levels of SIRT3,β-catenin and PPARγprotein were compared.Results:In the AMI group,the shape of the cells changed,the arrangement was disordered,and inflammatory infiltration and cell necrosis occurred.The morphology and arrangement of the cells in the resveratrol group were restored,and cell necrosis was significantly reduced.The levels of serum cTnT,CK-MB,IL-6 and TNF-αin the AMI group were significantly higher than those in the sham operation group(P<0.05).Serum cTnT,CK-MB,IL-6 and TNF-αlevels in the resveratrol group were significantly lower than those in the AMI group(P<0.05).The apoptosis rate of myocardial cells in the AMI group was significantly higher than that in the sham operation group(P<0.05).The apoptosis rate of cardiomyocytes in the resveratrol group was significantly lower than that in the AMI group(P<0.05).The levels of SIRT3,β-catenin and PPARγin myocardial tissue of the AMI group were higher than those in the sham operation group(P<0.05).The levels of SIRT3,β-catenin and PPARγin myocardial tissue of rats in the resveratrol group were significantly lower than those in the AMI group(P<0.05).Conclusion:Resveratrol relieves myocardial apoptosis induced by AMI by inhibiting SIRT3/β-catenin/PPARγpathway,and reduces inflammation to improve myocardial function.
