不同保存期的磷酸盐缓冲液(PBS)对鼠疫F1Ag绵羊红血球测定效果评价

Evaluation of phosphate buffer solution(PBS)for the determination of plague F1Ag sheep red blood cells

目的评价不同保存期磷酸盐缓冲液(phosphate buffer solution,PBS)对鼠疫F1Ag绵羊红血球的测定效果。方法对2019年和2020年两个不同时间段制备的PBS分别稀释鼠疫F1Ag至最适浓度(50~100μg/ml),按其F1Ag绵羊红血球操作流程进行制备,测定其效价,观察不同保存期的PBS对其测定效价的影响。结果2019年和2020年的PBS对制备的绵羊红血球,采用试管法从1∶100至1∶204800进行稀释,测定其效价结果发现不同批次的PBS对其制备的鼠疫F1Ag绵羊红血球的滴度均达到1∶50000倍以上,最高滴度可达1∶200000(可达F1Ag诊断试剂盒1∶50000的诊断标准)。结论不同保存期(保存一年和现场制备)的PBS对制备的鼠疫F1Ag绵羊红血球测定结果无影响。

Objective To evaluate the efficacy of phosphate buffer solution(PBS)at different storage periods for the determination of plague F1 Ag sheep red blood cells.Methods PBS prepared at two different periods in 2019 and 2020 was diluted to the optimal concentration(50-100μg/ml)of plague F1 Ag,respectively,and prepared according to the operation procedure of F1 Ag sheep red blood cell.The titer was determined,and the effect of PBS at different storage periods on the titer was observed.Results The potency of PBS from 2019 and 2020 was determined on prepared sheep red blood cells,diluted from 1∶100 to 1∶204800 using the test tube method,and the results found that different batches of PBS reached titers of more than 1∶50000 times against their prepared plague F1 Ag sheep red blood cells,with the highest titers reaching 1∶200000(Diagnostic criteria of 1∶50000 for F1 Ag diagnostic kit can be achieved).Conclusion PBS with different storage periods(one year and field preparation)had no effect on the determination results of the prepared plague F1 Ag sheep red blood cells assay.
