摘要
通过Suzuki偶联反应将给电子体4,4’-二甲氧基三苯胺连接在喹喔啉基上,制备得到发射红色荧光的化合物BMeQTF,并通过核磁共振氢谱和质谱对其结构进行了表征。利用Gaussian 09软件理论模拟了BMeQTF的构型和电子云分布。进一步,采用纳米沉淀法将其与两亲性的PLGA-PEG自组装得到BMeQTF纳米颗粒分散液。光谱性能测试显示其发射波长位于635 nm,且粒径约110 nm。将其用于小鼠成纤维L929细胞荧光成像,能够在细胞内看到明显的红色荧光。上述结果表明BMeQTF纳米颗粒具有亲水性和生物相容性的特点,有望作为荧光染料在生物成像和疾病示踪等方面受到青睐。
A novel emitting BMeQTF was prepared by Suzuki coupling reaction with quinoxaline and 4,4’-dimethoxytriphenylamine with strong electron-donated ability. Its structure was characterized by nuclear magnetic resonance spectroscopy(1 H NMR) and mass spectrometry(MS). The optimal configuration and electron cloud distribution of BMeQTF were observed by theoretical simulation with Gaussian 09. Nano-precipitation method was used to obtain dispersed BMeQTF nanoparticles in water with amphiphilic PLGA-PEG. Spectral performance test of BMeQTF nanoparticles were showed that the emission wavelength was located at 635 nm, and the particle size was about 110 nm in water. When applied to the fluorescence imaging of mouse fibroblast L929 cells, the visible red fluorescence could be seen in the cells. These results indicate that BMEQTF nanoparticles had the characteristics of hydrophilicity and biocompatibility, and were expected to be favored as a fluorescent dye in biological imaging and disease tracer.
