宫颈癌患者HMGA1、HMGB1表达水平与高危型HPV感染的关系及临床意义 被引量:7

The relationship between the expression levels of HMGA1 and HMGB1 in patients with cervical cancer and high⁃risk HPV infection and its clinical significance

摘要 目的探究宫颈癌患者高迁移率族蛋白A1(HMGA1)、高迁移率族蛋白B1(HMGB1)表达水平与高危型人乳头瘤病毒(HPV)感染的关系及临床意义。方法选取2017年8月至2020年8月南方医科大学附属小榄医院收治的200例宫颈癌患者作为研究组,另选取同期在本院体检健康女性就诊者50名作为对照组,研究组患者根据HPV感染情况分为高危组和非高危组。采用免疫组织化学法检测宫颈组织中HMGA1、HMGB1表达情况,采用PCR+膜杂交法检测HPV感染情况,采用酶联免疫吸附法(ELISA)检测血清中HMGA1、HMGB1水平。结果研究组宫颈组织中HMGA1、HMGB1高表达者明显多于对照组,差异有统计学意义(P<0.05);高危组患者组织中HMGA1、HMGB1高表达患者均明显多于低表达患者,差异有统计学意义(P<0.05);高危组患者血清中HMGA1、HMGB1高表达患者均明显多于低表达患者,差异有统计学意义(P<0.05);血清HMGA1、HMGB1联合诊断AUC为0.956(95%CI:0.878~0.999),高于单项(P<0.05)。宫颈癌患者组织中HMGA1、HMGB1水平与患者年龄、产次和肿瘤病理类型无关(P>0.05),与患者肿瘤分化程度、淋巴结转移及肿瘤FIGO分期相关(P<0.05)。结论宫颈患者组织中HMGA1、HMGB1水平明显升高,与患者肿瘤分化程度、淋巴结转移及肿瘤FIGO分期相关,且能够对宫颈癌合并高危型HPV感染做出早期诊断。 Objective To investigate the relationship between high mobility group protein A1(HMGA1),high mobility group protein B1(HMGB1)and high⁃risk human papillomavirus(HPV)infection in patients with cervical cancer and its clinical significance.Methods 200 cases of cervical cancer patients ad⁃mitted to Xiaolan Hospital affiliated to Southern Medical University from August 2017 to August 2020 were se⁃lected as the study group,and 50 healthy female patients who received physical examination in our hospital during the same period were selected as the control group.The patients in the study group were divided into high⁃risk group and non⁃high⁃risk group according to HPV infection.Immunohistochemistry was used to detect the expression of HMGA1 and HMGB1 in cervical tissues,PCR+membrane hybridization was used to detect HPV infection,and ELISA was used to detect the levels of HMGA1 and HMGB1 in serum.Results The high expression of HMGA1 and HMGB1 in cervical tissues of study group was significantly higher than that of control group,the difference was statistically significant(P<0.05).The high expression of HMGA1 and HMGB1 in the high⁃risk group was significantly higher than that in the low expression group(P<0.05);The high expression of HMGA1 and HMGB1 in the high⁃risk group was significantly higher than that in the low expression group,the difference was statistically significant(P<0.05);The AUC of HMGA1 and HMGB1 com⁃bined diagnosis was 0.956(95%CI:0.788⁃0.999),which was higher than that of single diagnosis(P<0.05).The levels of HMGA1 and HMGB1 in cervical cancer patients had no correlation with age,stage and patholog⁃ical type of tumor(P>0.05),but were correlated with tumor differentiation,lymph node metastasis and tumor FIGO stage(P<0.05).Conclusion The levels of HMGA1 and HMGB1 in cervical tissues were significantly increased,which were correlated with the degree of tumor differentiation,lymph node metastasis and tumor FIGO stage,and could be used for early diagnosis of cervical cancer complicated with high⁃risk HPV infection.
