慢性心力衰竭患者血清LncRNA生长抑制特异性转录本5表达与心肌重构及心功能的相关性研究 被引量:3

摘要 目的:探究长链非编码RNA生长抑制特异性转录本5(LncRNA GAS5)在慢性心力衰竭(CHF)患者的表达水平及与心肌重构、心功能的相关性。方法:选择本院2018年9月至2020年7月,收治的141例CHF患者为心力衰竭组;并以本院同时间段内的149例健康体检者为对照组。分析两组一般资料;实时荧光定量PCR(qRT-PCR)法检测两组血清中LncRNA GAS5表达水平;酶联免疫吸附试验(ELISA)法检测TNF-α、IL-10水平;比较两组心肌重构指标左心室重量(LVM)、LVEF水平;分析CHF患者血清LncRNA GAS5水平、LVM、LVEF与TNF-α、IL-10及LncRNA GAS5水平与LVM、LVEF的关系;Logistic回归分析CHF的影响因素。结果:心力衰竭组患者血清TNF-α及LVM水平明显高于对照组(P<0.05),血清LncRNA GAS5、IL-10、LVEF水平明显低于对照组(P<0.05);CHF患者血清LncRNA GAS5、LVEF与TNF-α水平,LVM与LncRNA GAS5、IL-10水平均呈负相关(P<0.05);LVM与TNF-α水平,LncRNA GAS5、LVEF与IL-10水平,LncRNA GAS5与LVEF水平均呈正相关(P<0.05);TNF-α、LVM为影响CHF发生的危险因素(P<0.05),LncRNA GAS5、LVEF是影响CHF发生的保护因素(P<0.05)。结论:LncRNA GAS5在CHF患者血清中呈低表达,与TNF-α、IL-10呈一定相关性,三者均可能在心肌重构、心功能变化中发挥作用。 Objective:To investigate the expression level of long non-coding RNA growth arrest specific transcript 5(LncRNA GAS5)in patients with chronic heart failure(CHF)and its correlations with myocardial remodeling and cardiac function.Methods:141 CHF patients diagnosed and treated in our hospital from September 2018 to July 2020 were selected as heart failure group;149 healthy persons in the same period of time were taken as the control group.The general data of the two groups were compared and analyzed;the levels of serum LncRNA GAS5 were measured by quantitative real-time PCR(qRT-PCR);the levels of serum TNF-αand IL-10 were measured by enzyme linked immunosorbent assay(ELISA);myocardial remodeling indexes left ventricular mass(LVM)and LVEF were compared between the two groups;the relationships between serum LncRNA GAS5 level,LVM,LVEF and TNF-α,IL-10,and the relationships between serum LncRNA GAS5 level and LVM,LVEF were analyzed;and the influencing factors of CHF were analyzed by Logistic regression.Results:The levels of serum TNF-αand LVM in heart failure group were significantly higher than those in control group(P<0.05),while the expression levels of serum LncRNA GAS5,IL-10 and LVEF were significantly lower than those of the control group(P<0.05);there were negative correlations between serum LncRNA GAS5,LVEF and TNF-αlevel,LVM and LncRNA GAS5,IL-10 level(P<0.05);there were positive correlations between LVM and TNF-αlevel,LncRNA GAS5,LVEF and IL-10 levels,LncRNA GAS5 and LVEF level(P<0.05),TNF-αand LVM were the risk factors of CHF(P<0.05),LncRNA GAS5 and LVEF were the protective factors of CHF(P<0.05).Conclusions:LncRNA GAS5 is low expressed in serum of CHF patients,and is correlated with TNF-αand IL-10,.and all of them may play a role in myocardial remodeling and cardiac function changes.
