摘要
目的探讨通痹胶囊缓解大鼠炎性痛的作用机制。方法以弗氏完全佐剂制备大鼠炎性痛模型,通痹胶囊(375、750、1500 mg·kg^(-1))灌胃治疗,观察大鼠体质量、爪水肿、机械缩足反射阈值及热缩足反射阈值的变化;采用酶联免疫吸附(ELISA)法检测大鼠脊髓组织中肿瘤坏死因子-α(TNF-α)、白细胞介素-1(IL-1)、白细胞介素-6(IL-6)的表达水平;蛋白免疫印迹法检测大鼠脊髓组织中NF-κB/P65、IκBα、IKKβ的蛋白表达。结果通痹胶囊能够明显升高弗氏完全佐剂所致炎性痛大鼠机械缩足反射阈值及热缩足反射阈值(P<0.05或P<0.01);降低大鼠脊髓组织中TNF-α、IL-1、IL-6的表达水平,抑制NF-κB/P65、IκBα、IKKβ的蛋白表达(P<0.05或P<0.01)。结论通痹胶囊具有明显的抗炎镇痛作用,其作用机制可能为抑制NF-κB信号通路的激活。
Objective To determine the mechanism of Tongbi capsules in relieving inflammatory pain in rats.Methods The inflammatory pain model of rat was prepared with Freund’s complete adjuvant,and the model rats were treated with Tongbi capsules(375,750,and 1500 mg·kg^(-1))by gavage.The changes in the weight,paw edema,mechanical withdrawal threshold and thermal withdrawal latency of the rats were observed.The expression levels of tumor necrosis factor-α(TNF-α),interleukin-1(IL-1)and interleukin-6(IL-6)in the spinal cord of rats were detected by ELISA.The expression of NF-κB/P65,IκBαand IKKβin the spinal cord of rats were detected by Western blot.Results Tongbi capsules significantly increased the mechanical withdrawal threshold and thermal withdrawal latency of the inflammatory pain in the model rats(P<0.05 or P<0.01).The expression levels of TNF-α,IL-1,IL-6 and NF-κB/P65,IκBα,IKKβwere decreased in the spinal cord tissue(P<0.05 or P<0.01).Conclusion Tongbi capsules have obvious anti-inflammatory and analgesic effect,whose mechanism may be inhibiting the activation of NF-κB signaling pathway.
