摘要
目的研究人参皂苷Rh_(3)联合多西他赛对前列腺癌(PCa)细胞系PC3细胞增殖、迁移和侵袭的影响。方法用WST-1法分别测人参皂苷Rh_(3)和多西他赛不同浓度处理24 h PC3肿瘤细胞的活力;人参皂苷Rh_(3)和多西他赛分别单独和联合处理PC3细胞后,用细胞划痕方法测PC3肿瘤细胞的迁移能力;Transwell法检测PC3肿瘤细胞的侵袭能力;Western blot法测PC3肿瘤细胞EMT相关标记蛋白E-cadherin和N-cadherin的表达水平。结果 不同浓度人参皂苷Rh_(3)和多西他赛处理PC3肿瘤细胞24 h,细胞活力不断下降,呈剂量依赖性。人参皂苷Rh_(3)(10 μmol·L^(-1))和多西他赛(2 nmol·L^(-1))处理24 h和48 h,均能显著抑制PC3细胞的迁移(P<0.05),但人参皂苷Rh_(3)联合多西他赛抑制PC3细胞迁移效果更明显(P<0.01);人参皂苷Rh_(3)和多西他赛干预24 h后均能显著抑制PC3细胞侵袭(P<0.05),两者联合作用效果更好(P<0.01)。人参皂苷Rh_(3)和多西他赛均能促进E-cadherin的表达,并抑制N-cadherin表达(P<0.05)。结论 人参皂苷Rh_(3)联合多西他赛抑制PC3肿瘤细胞的增殖、迁移和侵袭,可能是通过介导上皮-间质转化发挥作用。
Objective To determine the effect of ginsenoside Rh_(3) combined with docetaxel on the proliferation,migration and invasion of PC3 tumor cells.Methods WST-1 method was used to measure the proliferation of PC3 cells treated with different concentrations of ginsenoside Rh_(3) or docetaxel for 24 hours.After PC3 cells were treated with Rh_(3) and docetaxel alone or in combination,the cell wound healing test was used to determine the migration of PC3 cells.The Transwell method was used to test the invasion of PC3 cells.The protein expression levels of E-cadherin and N-cadherin related to EMT in the PC3 cells were measured by Western blot.Results PC3 cells were treated with different concentrations of ginsenoside Rh_(3) and docetaxel for 24 hours,and the cell activity decreased in a dose-dependent manner.Ginsenoside Rh_(3)(10μmol·L^(-1)) and docetaxel(2 nmol·L^(-1)) respectively inhibited the migration of PC3 cells for 24 hours and 48 hours(P<0.05)respectively;but combination of ginsenoside Rh_(3) with docetaxel worked better(P<0.01).Both ginsenoside Rh_(3) and docetaxel treatment inhibited the invasion of PC3 cells(P<0.05),but the combination worked better(P<0.01).Both ginsenoside Rh_(3) and docetaxel promoted the protein expression of E-cadherin and inhibited the expression of N-cadherin(P<0.05).Conclusion Both ginsenoside Rh_(3) and docetaxel inhibit the proliferation,migration and invasion of PC3 cells,possibly by mediating the epithelial-mesenchymal transition.
