黄芪增益方对小鼠免疫功能的影响

Immunomodulatory effect of Huangqi Zengyi formula on mice

目的探讨黄芪增益方对小鼠免疫功能的保健作用。方法小鼠分别经口灌胃黄芪增益方、匹多莫德3周后,用环磷酰胺制备免疫低下模型小鼠,继续灌胃黄芪增益方、匹多莫德1周,测定脾脏、胸腺指数观察药物对小鼠免疫器官的影响。采用碳廓清法检测单核-巨噬细胞功能,迟发型变态反应、淋巴细胞转化实验测定药物对小鼠免疫功能的影响,HE染色法检测模型小鼠脾脏、胸腺、胸骨骨髓的病理,ELASA法检测小鼠血清中免疫球蛋白A(lgA)、白介素-4(IL-4)。结果与模型组、阳性对照组比较,黄芪增益方可显著提高免疫抑制小鼠外周血白细胞、淋巴细胞、嗜中性白细胞数(P<0.05,P<0.01);提高胸腺指数和脾脏指数(P<0.01);增强淋巴细胞增殖能力(P<0.05);改善免疫器官与胸骨骨髓的免疫功能;显著上调血清lgA及细胞因子IL-4;提高小鼠的单核-巨噬细胞吞噬能力(P<0.05);抑制小鼠迟发型变态反应的耳肿胀系数(P<0.05)。结论黄芪增益方可改善小鼠免疫状态,增强免疫功能。

Objective To dertermine the prtective effect of Huangqi Zengyi formula on the immune function of mice.Methods After 3 weeks of oral administration of Huangqi Zengyi formula and pidomod,immunocompromised mouse models were prepared with cyclophosphamide.After another week of oral administration of Huangqi Zengyi formula and pidomod,the spleen and thymus indexes were measured to determine the effect of the drugs on the immune organs of these mice.Carbon clearance assay was used to detect the function of monocyte macrophage,delayed allergy and lymphocyte transformation test were used to determine the effect of drugs on the immune function of the mice.The pathological changes in the spleen,thymus and sternal bone marrow of the model mice were detected by HE staining.IGA and IL-4 in the serum of the mice were detected by ELASA.Results Compared with the model group and the positive control group,Huangqi Zengyi formula significantly increased the number of leukocytes,lymphocytes and neutrophils in the peripheral blood of immunosuppressed mice(P<0.05,P<0.01);increased the thymus and spleen indexes(P<0.01);enhanced the lymphocyte proliferation(P<0.05);improve the immune function of immune organs and sternal bone marrow;up-regulated the serum LGA and cytokine IL-4;increased the phagocytosis ability of the monocytes and the macrophages(P<0.05);and inhibited the ear swelling in mice with delayed allergic reaction in mice(P<0.05).Conclusion Huangqi Zengyi formula can improve the immunity and enhance the immune function of mice.
