摘要 目的研究不同产地牛蒡根药材的高效液相色谱(HPLC)指纹图谱,为其质量控制提供依据。方法采用HPLC法测定湖北、安徽、江西、辽宁、山东等9个产地的14批市售牛蒡根药材,色谱条件为安捷伦ZORBAX SB-C_(18)(250 mm×4.6 mm,5 μm)色谱柱,乙腈-0.04%磷酸溶液为流动相进行梯度洗脱,检测波长330 nm,流速 1.0 mL·min^(-1),柱温35℃,检测时间60 min。结果牛蒡根提取物中所含成分的各色谱峰均得到了有效分离,在测定的14批药材中,以3,5-二咖啡酰奎宁酸对照品为参照峰建立了牛蒡根的HPLC指纹图谱共有模式图,不同产地的样品相似度均在0.9以上,有11个共有峰,确定为特征指纹峰。结论该HPLC 指纹图谱具有可靠性,可为进一步研究制订牛蒡根的质量标准提供依据。 Objective To establish the HPLC fingerprints of the extract of Arctium lappa L.root from different origins,and to provide a basis for the identification and quality control of Arctium lappa L.root.Methods HPLC was used to determine 14 batches of commercially available Arctium lappa L.root from 9 areas in Hubei,Anhui,Jiangxi,Liaoning,Shandong,and other provinces.Agilent ZORBAX SB-C_(18)(250 mm×4.6 mm,5μm)chromatographic column was used.Acetonitrile-0.04%phosphoric acid solution was used as the mobile phase for gradient elution,the detection wavelength was 330 nm,the flow rate was 1.0 mL·min^(-1),the column temperature was 35℃,and the detection time was 60 min.Results The chromatographic peaks of the components in the extract of Arctium lappa L.root were well separated.Among the 14 batches of medicinal materials measured,3,5-dicaffepyridinequinic acid was used as the reference peak to establish a common pattern of HPLC fingerprints of the extract of Arctium lappa L.root.The similarity of fingerprint of all samples was above 0.9,and 11 common peaks were found,which were identified as characteristic fingerprint peaks.Conclusion The HPLC fingerprint established by this method is reliable,which provides a basis for further research and quality standard of extract of Arctium lappa L.root.
