蒙药山沉香抗心肌缺血及其作用机制研究 被引量:3

摘要 目的:探讨蒙药山沉香抗心肌缺血的作用及可能机制,从而为该药后续的开发研究提供科学依据,也为其临床应用奠定部分基础。方法:Wistar大鼠分别给予山沉香原粉、醇提物、挥发油、醇提物+挥发油,采用皮下注射异丙肾上腺素(ISO)造成急性心肌缺血模型,末次给药后观察心电图变化,HE染色观察大鼠心肌组织病理变化,RT-PCR法检测Bcl-2、Bax基因的表达。结果:心电监测结果表明,山沉香各处理组均对ISO所致急性心肌缺血具有治疗作用,其中醇提物组治疗作用最明显,能够显著改善大鼠心电图ST段抬高(P<0.05);大鼠心肌组织HE染色结果表明,与模型组相比,山沉香各处理组显微镜下观察均病变减轻,其中醇提物组与原粉组心肌组织病变最轻;RT-PCR结果表明,除挥发油组外,山沉香其余各处理组均能下调促凋亡因子Bax的表达,上调抗凋亡因子Bcl-2的表达,Bcl-2/Bax比值显著高于模型组。其中醇提物组明显优于其他各组(P<0.01)。结论:蒙药山沉香能够对ISO所致急性心肌缺血形成保护作用,综合比较显示山沉香醇提物作用最显著。山沉香抗心肌缺血的作用机制可能是通过上调抗凋亡因子Bcl-2的表达,下调促凋亡因子Bax的表达,抑制细胞凋亡发挥作用。 Objective:To explore the anti-myocardial ischemia effect and possible mechanism of the Mongolia medi-cine“Syringa pinnatifolia Hemsl”,so as to provide the scientific basis for its subsequent researches,and also lay a partial foundation for its clinical application.Methods:Wistar rats were administered with the powder,alcohol extract,volatile oil,alcohol extract+volatile oil of the Mongolia medicine“Syringa pinnatifolia Hemsl”respectively,and isoproterenol(ISO)was used to create the acute myocardial ischemia model.After the last administration,electrocardiogram(ECG)changes were ob-served,pathological changes of myocardial tissue were investigated by HE staining,and RT-PCR was used to detect the ex-pression of Bcl-2 and Bax genes.Results:The results of ECG monitoring showed that all the treatment groups of the Mongo-lia medicine“Syringa pinnatifolia Hemsl”have therapeutic effect on ISO-induced acute myocardial ischemia,and the alco-hol extract group had the most obvious therapeutic effect,which could significantly improve the ST segment elevation of ECG in rats(P<0.05).The results of HE staining of rat myocardium tissue showed that,compared with the model group,the patho-logical changes in each treatment group were reduced under microscope,and the alcohol extract group and original powder group have the least pathological changes.RT-PCR results showed that,except the volatile oil group,the expression of pro apoptotic factor Bax could be down-regulated and the expression of anti-apoptotic factor Bcl-2 could be up-regulated in all the other treatment groups,and the ratio of Bcl-2/Bax was significantly higher than that in the model group.The alcohol ex-tract group is significantly better than the other groups(P<0.001).Conclusion:The Mongolia medicine“Syringa pinnatifo-lia Hemsl”can protect against ISO-induced acute myocardial ischemia,and comprehensive comparison shows that the alco-hol extract has the most significant effect.The anti-myocardial ischemia mechanism may be that it inhibits apoptosis by up regulating the expression of anti-apoptotic factor Bcl-2 and down-regulating the expression of pro-apoptotic factor Bax.
