荞麦蜜提取液对西方蜜蜂存活及Vg和Sir2基因表达量的影响 被引量:1

摘要 为探讨荞麦蜜提取液对西方蜜蜂存活及寿命基因表达量的影响,采用4种荞麦蜜提取液对西方蜜蜂成年工蜂进行饲喂,分析各提取液对不同日龄成年工蜂的存活率、Vg和Sir2基因表达量。结果表明,荞麦蜜正丁醇提取液饲喂的成年工蜂存活率最高,35日龄时存活率为56.27%。取食3种荞麦蜜提取液(乙酸乙酯、正丁醇、乙腈)的成年工蜂存活率在10~25日龄期间相对较高,25日龄后存活率急剧下降。3种荞麦蜜提取液饲喂的成年工蜂Vg相对表达量呈现先上升后下降的趋势,在27日龄达到峰值;Sir2相对表达量随日龄增加呈现逐渐下降的趋势。说明在成年工蜂10~25日龄采取荞麦蜜正丁醇提取液进行饲喂,可维持成工年蜂较高的存活率,有利于蜂群繁育。Vg相对表达量可能受到蜜蜂行为发育中保幼激素滴度水平的影响。 In order to investigate the effects of buckwheat honey extract on the survival and longevity gene expression of Apis mellifera,the survival rate,Vg and Sir2 gene expression of adult worker bees of different days of age were analyzed by feeding them with four kinds of buckwheat honey extracts.The results showed that adult worker bees fed with n-butanol extract of buckwheat honey had the highest survival rate,and the survival rate on 35 days of age was 56.27%.The survival rate of adult worker bees fed with three kinds of buckwheat honey extracts(ethyl acetate,n-butanol,acetonitrile)was relatively high during the period of 10 days of age to 25 days of age,and the survival rate decreased sharply after 25 days of age.The relative expression of Vg in adult worker bees fed with three kinds of buckwheat honey extracts showed a trend of increasing first and then decreasing,and reached the peak on 27 days of age;the relative expression of Sir2 showed a gradually decreasing trend with the increase of days.These results indicate that adult worker bees fed with n-butanol extract of buckwheat honey from 10 days of age to 25 days of age could maintain a higher survival rate,which was beneficial to the breeding of A.mellifera.The relative expression of Vg may be affected by the titer level of juvenile hormone during behavioral development of A.mellifera.
