摘要 目的研究蛇床子素对人舌鳞癌Tca8113细胞增殖、凋亡和自噬的影响,并探讨其可能的作用机制。方法体外培养Tca8113细胞,并将细胞分为不加药的对照组和分别加入40、80、120、160μmol/L蛇床子素的实验组。用四甲基偶氮唑盐实验和集落形成实验检测蛇床子素对Tca8113细胞的增殖作用,通过Hoechst33342染色法和流式细胞仪检测蛇床子素对人舌鳞癌Tca8113细胞24 h凋亡情况,Western blot检测Bcl-2、Bax、活化的半胱氨酸蛋白酶3及LC3、P62的表达情况。结果蛇床子素可以明显抑制Tca8113细胞增殖,且药物对细胞的抑制率呈浓度依赖性;细胞集落形成能力降低;Hoechst33342染色和流式细胞术结果显示,蛇床子素呈浓度依赖性诱导细胞死亡;Western blot结果显示,蛇床子素能上调Bax和活化的半胱氨酸蛋白酶-3蛋白表达,下调Bcl-2蛋白表达。同时增加LC3Ⅱ、P62的表达,降低LC3Ⅰ的表达。结论蛇床子素能抑制人舌鳞癌Tca8113细胞增殖,其机制可能与促进细胞凋亡并抑制细胞自噬流、自噬途径损伤而抑制细胞自噬有关。 Objective To investigate the effects of osthole on the proliferation,apoptosis,and autophagy of human tongue cancer Tca8113 cells and explore its possible mechanism of action.Methods Tca8113 cells were cultured in vitro and divided into a control group without drugs and the experimental groups with 40,80,120,and 160μmol/L osthole.The inhibitory effect of osthole on the proliferation of Tca8113 cells was detected by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay and colony formation assay.Hoechst33342 staining method and annexin V-FITC/propidium iodide method were employed to detect the effect of osthole on the apoptosis of Tca8113 cells within 24 hours.Western blot was performed to detect the expression of apoptosis-related proteins(Bcl-2,Bax,and cleaved caspase-3)and autophagy-related proteins(LC3 and p62)in Tca8113 cells exposed to osthole.Results Osthole significantly inhibited the proliferation and induced the apoptosis of Tca8113 cells in a concentration-dependent manner,and it reduced the cell colony formation.Western blot results showed that osthole could up-regulate the expression of Bax and cleaved caspase-3 and down-regulate that of Bcl-2.At the same time,it increased the expression of LC3Ⅱand P62 and reduced that of LC3Ⅰ.Conclusion Osthole may inhibit the proliferation of Tca8113 cells by promoting cell apoptosis and blocking autophagy flow to inhibit autophagy.
