2型糖尿病db/db小鼠血管内皮细胞原代培养和生长曲线检测

Primary Culture and Growth Curve Detection of Vascular Endothelial Cells in Type 2 Diabetic db/db mice

目的探讨糖尿病小鼠血管组织块血管内皮细胞原代培养方法,并检测其生长曲线。方法分离2型糖尿病db/db小鼠胸主动脉和腹主动脉,采用组织块贴壁法获得血管内皮细胞,通过传代对细胞进纯化,并对细胞进行生长曲线检测。结果血管组织块培养可快速高效获得的血管内皮细胞,并且细胞特性稳定。结论成功建立了2型糖尿病db/db小鼠血管内皮细胞原代培养方法,获得的种子细胞可进行血管相关疾病研究。

Objective To explore the primary culture method of vascular endothelial cells of diabetic db/db mice and detect its growth curve.Method Vascular endothelial cells were obtained from thoracic aorta and abdominal aorta of type 2 diabetic db/db mice by tissue attachment method.The cells were purified by subculture and their growth curves were detected.Result The vascular endothelial cells were obtained rapidly and efficiently had stable cell characteristics.Conclusion The primary culture method of vascular endothelial cells in type 2 diabetic db/db mice had been successfully established,and the seed cells obtained could be used for the study of vascular related diseases.
