19个短串联重复序列基因座在河南汉族人群中的遗传多态性分析 被引量:1

摘要 目的调查19个常染色体短串联重复序列(STR)基因座在河南汉族人群中的遗传多态性,对其在河南汉族人群法医学中的应用效能进行验证。方法随机选取河南事缘法医物证司法鉴定所2019—2020年日常接收案件中1000个互不相关的河南汉族健康个体的样本。采用Goldeneye TM 20A免提取直扩试剂盒对19个常染色体STR基因座进行扩增及检测,统计其遗传多态性。结果在19个STR基因座中共检出245个等位基因和980种基因型。等位基因的频率为0.001~0.530,分布均符合Hardy-Weinberg平衡定律,累积个体识别能力(DP)为(1~1.81499)×10^(-23),累积非父排除概率(PE)为0.999999988。结论本研究中19个常染色体STR基因座在河南汉族人群中具有较高的个体DP和亲权鉴定能力。 Objective To investigate the genetic polymorphisms of 19 autosomal short tandem repeat(STR)loci in population of Henan Han,and to verify their effectiveness in forensic medicine in Henan Han population.Methods A random sample of 1000 unrelated healthy individuals of Henan Han nationality in the daily cases received by Henan Shiyuan Institute of Forensic Material Evidence from 2019 to 2020 was selected.The Goldeneye TM 20A extraction-free direct amplification kit was used to amplify and detect 19 autosomal STR loci,and count their genetic polymorphisms.Results A total of 245 alleles and 980 genotypes were detected in 19 STR loci.The frequency of alleles ranged from 0.001 to 0.530,and the distribution conformed to the law of Hardy-Weinberg equilibrium.Cumulative individual discrimination power(DP)was(1 to 1.81499)×10^(-23),and the cumulative non-paternal probability of exclusion(PE)rate was 0.999999988.Conclusion The 19 autosomal STR loci studied in this study have high individual DP and parental identification in Henan Han population.
