鼠巨细胞病毒升高血压并引起NLRP3炎症小体激活 被引量:1

摘要 目的通过采用腹腔感染鼠巨细胞病毒(MCMV)构建小鼠高血压模型,探究MCMV感染引发的高血压与炎症小体之间的关系。方法将7月龄雄性C57BL/6J小鼠分为腹腔注射生理盐水组(n=10)与腹腔注射MCMV实验组(MCMV组,n=10)。使用颈动脉测压法比较腹腔注射MCMV 5月后两组小鼠血压变化情况,采用Western Blot及免疫组织化学法检测血管中NOD样受体热蛋白结构域相关蛋白3(NOD-like receptor pyrin domain containing 3,NLRP3)、CARD凋亡相关微粒蛋白(Apoptosis-associated speck-like protein containing a CARD,ASC)、半胱氨酸天冬蛋白酶-1(Cysteinyl-aspartate specific protease-1,Caspase-1)、白介素18(Interleukin 18,IL-18)、白介素1β(Interleukin 1β,IL-1β)的蛋白表达情况。结果与对照组小鼠相比,MCMV组小鼠收缩压、舒张压与平均动脉压均显著升高,且小鼠主动脉组织中炎症小体相关蛋白NLRP3、ASC、Caspase-1、IL-18、IL-1β表达均显著增加。结论MCMV感染可能通过激活NLRP3等炎症小体引起小鼠血压升高。 Objective To explore the relationship between hypertension caused by MCMV infection and inflammasomes by constructing a mouse model of hypertension by using murine cytomegalovirus(MCMV)infection in the abdominal cavity.Methods Male C57BL/6J mice aged 7 months were divided into an intraperitoneal injection of normal saline group(n=10)and an intraperitoneal injection of MCMV experimental group(MCMV group,n=10).Using carotid artery manometry to compare the changes in blood pressure of the two groups of mice after 5 months of intraperitoneal injection of MCMV,Western Blot and immunohistochemistry were used to detect NODlike receptor pyrin domain containing 3(NLRP3)and CARD apoptosis-related microparticle protein(Apoptosis-associated speck-like protein containing 3)in blood vessels.a CARD,ASC),Cysteinyl-aspartate specific protease-1(Caspase-1),Interleukin 18(IL-18),Interleukin 1β(Interleukin 1β,IL-1β)protein expression.Results Compared with mice in the control group,the systolic blood pressure,diastolic blood pressure,and mean arterial pressure of the MCMV group increased significantly,and the inflammasome-related proteins NLRP3,ASC,Caspase-1,IL-18,The expression of IL-1βincreased significantly.Conclusion MCMV infection may increase blood pressure in mice by activating inflammasomes such as NLRP3.
