Expression of NTN4 in lung squamous carcinoma and its correlation with prognosis

目的研究肺鳞癌组织中神经导向因子4(NTN4)mRNA表达水平,并分析其在预测患者预后中的作用。方法Oncomine数据库查询NTN4基因在肺癌中的表达情况,并对肺鳞癌数据进行荟萃分析;GEPIA数据库进一步确认NTN4 mRNA在肺鳞癌组和癌旁正常组织中的存在状态,并分析其表达量高低与患者总体生存率的关联。RT-PCR检测肺鳞癌组织和癌旁的NTN4基因mRNA表达情况,并分析与临床参数的相关性。结果Oncomine数据库提示共有7项研究结果表明,NTN4在肺癌组织中比癌旁正常组织表达低,其水平在肺鳞癌中也明显下降;GEPIA数据库显示,NTN4低表达的肺鳞癌患者总体生存率(OS)明显高于NTN4高表达的肺鳞癌患者。89例肺鳞癌组织NTN4基因表达量显著低于癌旁对照,且其水平与临床分期、分化程度和淋巴结转移具有显著相关性。结论NTN4在肺癌尤其是肺鳞癌组织中表达量减少,且与患者总体生存率呈正相关。

Objective To study the level of neuroguidance factor 4(NTN4)mRNA in lung squamous carcinoma tissue and its role in predicting prognosis.Methods The expression of NTN4 gene in lung cancer was investigated by Oncomine database,and the data of lung squamous carcinoma were analyzed.The GEPIA database further confirmed the existence of NTN4 mRNA in the lung squamous carcinoma and normal tissues near the cancer,and analyzed the relationship between its expression level and the overall survival rate of the patient.RT-PCR was used to detect the expression of NTN4 gene in lung squamous cell carcinoma tissues and adjacent tissues,and the correlation with clinical parameters was analyzed.Results The Oncomine data⁃base showed that NTN4 had lower expression in lung cancer tissue than in the normal tissue next to cancer,and its level decreased significantly in lung squamous carcinoma.The GEPIA database shows that the overall survival rate(OS)of patients with lower expression of NTN4 is significantly higher than that of patients with higher expression of NTN4.The expression of NTN4 gene in 89 cases of lung squamous cell carcinomas was significantly lower than that in the adjacent control group,and its level was significantly correlated with clinical stage,differentiation degree and lymph node metastasis.Conclusion NTN4 expression have been decreased in lung cancer,especially in lung squamous carcinoma,and its level was positively correlated with overall survival rate.
