CD14^(+) HLA-DR^(low)髓源性抑制细胞在原发性肝癌患者中的研究

摘要 目的探索CD14^(+)HLA-D^(Rlow)髓源性抑制细胞(MDSC细胞)在原发性肝癌患者中分布情况及其临床意义,并与其他免疫细胞进行相关性分析。方法收集53例原发性肝癌患者外周血,BD CantoⅡ流式细胞仪器检测原发性肝癌患者外周血标本中CD14^(+)HLA-D^(Rlow)、CD3^(+)T细胞、CD4^(+)T细胞、CD8^(+)T细胞以及CD16^(+)CD56^(+)NK细胞表达,与其他免疫细胞进行相关性分析对发病机制进行研究。结果与31例健康者相比,53例原发性肝癌患者外周血中CD14^(+)HLA-D^(Rlow)细胞表达显著增加,其中CD14^(+)HLA-D^(Rlow)细胞高表达与原发性肝癌患者分期及预后显著相关。且比例与CD3^(+)T及CD8^(+)T细胞比例呈负相关,但与CD4^(+)T及CD16^(+)CD56^(+)NK细胞比例无相关。结论CD14^(+)HLA-D^(Rlow)细胞在原发性肝癌患者外周血高表达且与患者预后相关,可能与抑制CD3^(+)T细胞及CD8^(+)T细胞功能相关。 Objective To explore the expression and clinical significance of CD14^(+)HLA-D^(Rlow) myeloid-derived suppressor cells(MDSC cells)in primary liver cancer patients,and to analyze the correlation between MDSC cells and other lymphocyte subsets.Methods The frequency of CD14^(+)HLA-D^(Rlow),CD3^(+)T cells,CD4^(+)T cells,CD8^(+)T cells and CD16^(+)CD56^(+)NK cells in peripheral blood samples of 53 primary liver cancer patients were detected by BD CantoⅡflow cytometry,and the potential immune pathogenesis was investigated by correlation analysis with other lymphocyte subsets.Results The expression of CD14^(+)HLA-D^(Rlow) cells was significantly increased in the peripheral blood of 51 primary liver cancer patients.The high expression of CD14^(+)HLA-D^(Rlow)cells was significantly associated with the stage and the prognosis of primary liver cancer.And its proportion was negatively associated with the proportion of CD3^(+)T cells and CD8^(+)T cells,but not with the proportion of CD4^(+)T cells and CD16^(+)CD56^(+)NK cells.Conclusion The high expression of CD14^(+)HLA-D^(Rlow) cells in the peripheral blood of primary liver cancer patients is associated with the prognosis of liver cancer patients,which may be related to the inhibi⁃tion of CD3^(+)T and CD8^(+)T cells.
