左乙拉西坦对新生大鼠缺血缺氧性脑损伤的保护作用

摘要 目的研究左乙拉西坦通过调节核因子κB(nuclear factor kappa-B,NF-κB)对新生大鼠脑缺血缺氧性脑损伤的影响。方法将新生SD大鼠随机分为假手术组、模型组和实验组,每组5只。实验组在模型构建前48 h给予150 mg·kg^(-1)·d^(-1)左乙西拉坦,灌胃,模型组和假手术组给予生理盐水,灌胃。以Bederson法评估大鼠神经功能,以逆转录实时荧光定量聚合酶链反应(qRT-PCR)检测大鼠脑组织中caspase-3表达。以蛋白质印迹(Western blot)法检测大鼠脑组织中NF-κB表达水平。结果假手术组、模型组和实验组的神经Bederson评分分别为0,2.40±0.49和0.60±0.49;caspase-3相对表达量分别为1.02±0.13,1.63±0.29和1.22±0.09;NF-κB的相对表达量分别为1.01±0.17,2.57±0.28和1.52±0.20;模型组与假手术组相比,实验组与模型组相比,差异均有统计学意义(P<0.05,P<0.01)。结论左乙拉西坦通过下调NF-κB保护新生大鼠缺血缺氧性脑损伤。 Objective To study the effects of levetiracetam on the hypoxic-ischemic brain damage in neonatal rats by regulating nuclear factor-κB(NF-κB)pathway.Methods The neonatal SD rats were randomly divided into sham operation group,model group and experimental group,5 rats in each group.Rats in experimental group was given 150 mg·kg^(-1)·d^(-1) levetiracetam at 48 h before model construction by gavage.Rats in model group and sham operation group was given normal saline by gavage.The Bederson method was used to evaluate the neurological function of rats,real-time quantitative reverse transcription polymerase chain reaction(qRT-PCR)assay was conducted to detect the expression of caspase-3 in rat brain tissues.Western blot assay was used to detect the expression of NF-κB in rat brain tissues.Results The Bederson neurological scores in sham operation group,model group and treatment group were 0,2.40±0.49 and 0.60±0.49;the relative expressions of caspase-3 were 1.02±0.13,1.63±0.29 and 1.22±0.09;the relative expressions of NF-κB were 1.01±0.17,2.57±0.28 and 1.52±0.20.The differences between model group and sham operation group,as well as between experimental group and model group were statistically significant(P<0.05,P<0.01).Conclusion Levetiracetam protects neonatal rats from hypoxic-ischemic brain damage by down-regulating NF-κB.
