新型水解蛋清蛋白短肽对大鼠结肠炎症的保护作用 被引量:1

摘要 目的:通过酶解方法获得新型水解蛋清蛋白短肽(HEpep),探究其对细菌内毒素脂多糖(LPS)所致大鼠结肠炎症的保护作用。方法:将60只雄性SD大鼠随机分为空白组、模型组、HEpep高剂量组和低剂量组。HEpep高【2 g/(kg·d)】、低剂量组【1 g/(kg·d)】连续7 d灌胃给药,空白组和模型组采用生理盐水同步处理;第7天腹腔注射LPS 10 mg/kg,给药8 h后经腹主动脉取血并收集结肠组织;通过HE染色和TUNEL染色观察结肠上皮细胞凋亡和病理组织学变化;通过ELISA试剂盒检测血清、组织中炎症因子肿瘤坏死因子α(TNF-α)、白细胞介素-6(IL-6)水平变化;通过生化试剂盒检测血清、组织中氧化应激指标超氧化物歧化酶(SOD)、丙二醛(MDA)及过氧化氢酶(CAT)的变化。结果:HEpep能够显著缓解LPS所致大鼠结肠炎症反应,改善结肠组织病理性损伤和结肠上皮细胞凋亡,降低组织和血清中TNF-α和IL-6炎症因子水平,对LPS所致氧化应激损伤具有明显的保护作用。结论:HEpep可减轻LPS所致大鼠结肠炎症反应,其作用机制与抗炎、抗氧化及抗凋亡有关。 Objective:To investigate the protective effect of hydrolyzed egg white short chain polypeptide(HEpep)on LPS-induced colitis in rats.Method:60 male SD rats were randomly divided into 4 groups:control group,model group,and high or low-dose of HEpep group.Rats in HEpep groups were administrated orally for 7 days at 2 g/(kg·d)or 1 g/(kg·d),while the control and model group were treated with normal saline synchronously.On day 7,all rats were intraperitoneally injected with LPS of 10 mg/kg.8 h after administration,blood and colon tissues were collected.Apoptosis and pathological changes were observed by HE staining and TUNEL staining.The levels of inflammatory factors-αand interleukin-6 in serum and tissue were detected by ELISA.Superoxide dismutase(SOD),malondialdehyde(MDA)and catalase(CAT)in serum and tissues were tested.Results:HEpep significantly alleviated LPS-induced inflammatory response in colon tissues,relieved the pathological injury and apoptosis,reduced the levels of TNF-αand IL-6 and oxidative stress injury in tissues and serum.Conclusion:HEpep could alleviate the inflammatory response in colon tissues caused by LPS,and the mechanism is related to anti-inflammation,anti-oxidation and anti-apoptosis.
