摘要 目的研究Nesfatin-1对人胃癌AGS细胞增殖的影响及其可能的影响机制。方法将人胃癌AGS细胞分为Nesfatin-1组和对照组,分别用浓度为0.1、1、10、100 nmol/L的Nesfatin-1刺激胃癌AGS细胞,采用CCK-8法检测各组细胞的增殖情况。选择最佳浓度的Nesfatin-1来刺激AGS细胞,观察Rho激酶抑制剂(Y-27632)对胃癌AGS细胞增殖的影响;各组细胞内RhoA、ROCK-1蛋白表达采用Westem blotting法检测。结果Nesfatin-1呈浓度和时间依赖性促进胃癌AGS细胞的增殖(P<0.05),胃癌细胞内RhoA、ROCK-1蛋白表达明显升高(P<0.05),ROCK抑制剂Y-27632可阻滞Nesfatin-1促进胃癌AGS细胞的增殖(P<0.05),降低RhoA、ROCK-1蛋白表达(P<0.05)。结论Nesfatin-1可促进胃癌AGS细胞的增殖,其机制可能与其调节RhoA/ROCK-1信号通路有关。 Objective To investigate the effects and the underlying mechanisms of Nesfatin-1 on the proliferation of gastric cancer AGS cells.Methods AGS cells were divided into the Nesfatin-1 group and the control group.After treatment with different concentrations of Nesfatin-1,the proliferation of gastric cancer AGS cells were detected by CCK-8.AGS cells treated with optimal concentration of Nesfatin-1 and observed the effect of Rho kinase inhibitor(Y-27632)on Nesfatin-1 induced proliferation.The protein expression of RhoA and ROCK-1 in every group were detected by Western blotting.Results Nesfatin-1 could promote AGS cells proliferation in a dose and time-dependent manner(P<0.05).The expressions of RhoA and ROCK-1 protein were enhanced significantly after Nesfatin-1 intervention.However,ROCK inhibitor Y-27632 significantly inhibited Nesfatin-1 induced cell proliferation,and decreased the protein expressions of RhoA and ROCK-1.Conclusion Nesfatin-1 may promote gastric cancer AGS cells proliferation,and the mechanism may be through regulating RhoA/ROCK-1 signal pathway.
