丹参多糖对氟苯尼考导致的肉鸡生长抑制和肝功能损伤的缓解作用

摘要 为了探究丹参多糖对氟苯尼考导致肉鸡肝脏损伤的缓解效果,40只1日龄的肉鸡被随机分成4组,每组10只。0.15 g/L氟苯尼考通过连续5 d饮水给药诱导肉鸡肝脏损伤,同时给与5.00 g/L的丹参多糖进行治疗。于42日龄翅下静脉采血,并处死肉鸡采取肝脏。统计肉鸡的增重及肝脏指数;血细胞计数仪检测全血中各成分的含量;半自动生化分析仪检测雏鸡血清中AST、ALT、 MDA、NO、SOD和CAT的含量;ELISA检测雏鸡肝组织内细胞色素酶CYP1A1和CYP2H1的含量;Q-PCR检测肝组织中CYP1A1、CYP2H1、Nrf2、HO-1、NQO-1、UGT2A1和GSTT1 mRNA的转录水平。结果显示,与空白组相比,氟苯尼考能显著抑制肉鸡增重,显著提高肉鸡肝脏指数(P<0.05);氟苯尼考还引起肉鸡血液中RBC和HGB的显著下降以及血清中MDA和NO的显著升高(P<0.05),显著抑制肝脏中CYP1A1的表达及Nrf2和GSTT1 mRNA的转录水平;而丹参多糖能显著抑制氟苯尼考导致的肉鸡的增重下降和肝指数升高(P<0.05),恢复血液中RBC和HGB的含量,丹参多糖还能显著降低肉鸡血清中MDA和NO的含量,丹参多糖还能显著提高肝组织中CYP1A1、Nrf2和GSST1的mRNA转录水平的表达(P<0.05)。结果表明,到42日龄时,氟苯尼考还能抑制肉鸡增重,还有对血细胞和肝脏有轻微毒性作用,而丹参多糖能够有效的化解这种毒性,其作用方式可能是丹参多糖通过调节肝细胞内的氧化应激和肝脏药物代谢能力,而达到保护氟苯尼考导致的肝细胞损伤。 In order to explore the effect of Salvia miltiorrhiza polysaccharides(SMPs) on liver injury induced by florfenicol(FFC) in broilers, forty one-day-old broilers were randomly divided into four groups, ten broilers in each group.Liver injury was induced by drinking water containing 0.15 g/L FFC for 5 d, at the same time, 5.00 g/L SMPs were given for treatment.At 42 days of age, the blood was collected from the inferior wing vein, and then the broilers were killed to take the liver.The content of each component in whole blood was detected by blood cell counter;the contents of AST,AST,MDA,NO,SOD and CAT in serum were detected by semi-automatic biochemical analyzer;the contents of cytochrome enzymes CYP1 A1 and CYP2 H1 in liver tissue were detected by ELISA;the mRNA transcription levels of CYP1 A1,CYP2 H1,Nrf2,HO-1,NQO-1,UGT2 A1 and GSTT1 in liver tissue were detected by q-PCR.The results showed that compared with the control group, FFC significantly inhibited the weight gain of broilers and significantly increased the liver index of broilers(P<0.05);FFC also significantly decreased the contents of RBC and HGB in blood and significantly increase MDA and NO in serum(P<0.05),and significantly inhibited the expression of CYP1 A1,Nrf2 and GSTT1 in liver.SMPs could significantly inhibit the decrease of weight gain and the increase of liver index caused by FFC(P<0.05),it could restore the contents of RBC and HGB in blood(P<0.05),significantly reduce the contents of MDA and NO in serum,and significantly increase the mRNA transcription level of CYP1 A1,Nrf2 and GSTT1 in liver tissue(P<0.05).It can be concluded that FFC can inhibit the weight gain of broilers at 42 days of age,and it has slight toxic effect on blood cells and liver,SMPs can effectively reduce toxic effect of FFC and protect liver cell from damage caused by FFC via regulating oxidative stress and drug metabolism in liver cells.
