茶多酚通过NF-κB通路缓解LPS诱导的奶牛乳腺上皮细胞炎性反应 被引量:12

Tea polyphenols alleviate LPS-induced inflammation in bovine mammary epithelial cells via the NF-κB pathway

摘要 为了探讨茶多酚对脂多糖(lipopolysaccharide, LPS)诱导的奶牛乳腺上皮细胞炎性反应的影响,本试验使用LPS(1 mg/L)和或茶多酚(50,100,200 mg/L)处理奶牛乳腺上皮细胞(MAC-T),随后采用ELISA试剂盒、Western blot方法和实时荧光定量PCR方法检测NF-κB信号通路关键分子及下游靶基因的表达变化。结果显示,与LPS组相比,茶多酚+LPS组中IKK-β活性显著降低,IκB-α和NF-κB的磷酸化水平也显著降低。此外,茶多酚+LPS组中TNF-α、IL-6和IL-1βmRNA表达量也显著低于LPS组。结果表明,茶多酚可以通过抑制NF-κB炎性信号通路缓解LPS诱导的奶牛乳腺上皮细胞炎性反应。 To investigate the effect of tea polyphenols on inflammation of bovine mammary epithelial cells(BMEC) induced by lipopolysaccharide(LPS),BMEC were treated with LPS(1 mg/L) and/or tea polyphenols(50,100,200 mg/L).The ELISA kit, Western blot and real-time fluorescence quantitative PCR were used to detect the expression of key molecules and downstream target genes of NF-κB signaling pathway.The results showed that, compared with the LPS group, the activity of IKK-β as well as the phosphorylated IκB-α and NF-κB were significantly decreased in tea polyphenols + LPS group.In addition, the mRNA abundance of TNF-α,IL-6 and IL-1β were also significantly decreased in tea polyphenols + LPS group compared with the LPS group.These results indicated that tea polyphenols can alleviate the inflammation of BMEC induced by LPS by inhibiting the NF-κB pathway.
