摘要
为研究大鼠70%肝缺血再灌注(ischemia-reperfusion,IRI)合并部分肝切除损伤对机体氧化应激和炎症反应的影响,将48只SD大鼠随机分为假手术组和模型组,假手术组仅翻动肝叶,模型组肝中叶及左叶缺血30 min,再灌注前切除肝左叶,检测各组血清中ALT水平,氧化应激试剂盒检测血清中SOD、GSH-Px、MDA水平,ELISA法检测血清中TNF-α、IL-1β、IL-6、IL-10水平。结果显示,模型组SOD、GSH-Px水平于再灌注后6,24 h较假手术组均显著性降低,MDA水平显著性升高;模型组TNF-α、IL-1β、IL-6、IL-10水平于再灌注后24,72 h较假手术组均显著性升高。结果表明,大鼠70%肝缺血再灌注合并部分肝切除损伤可导致机体氧化应激和炎症反应的发生,其机制可能与抗氧化酶水平的下调及促炎因子表达增强有关。
The aim of this study is to determine the effect of 70%liver ischemia-reperfusion(IRI)combined with partial hepatectomy injury on oxidative stress and inflammation in rats.Forty-eight SD rats were randomly divided into sham operation group and model group.The sham operation group only turned the hepatic lobe.In the model group,the middle lobe and left lobe of the liver were ischemic for 30 min,and the left lobe of the liver was removed before reperfusion.The levels of ALT,SOD,GSH-Px and MDA in serum were detected by oxidative stress kit,and the levels of TNF-α,IL-1β,IL-6 and IL-10 in serum were detected by ELISA.The results showed that the levels of SOD and GSH-Px in the model group were significantly lower than those in the sham operation group at 6,24 h after reperfusion,while the levels of MDA in the model group were significantly higher than those in the sham operation group at 24,72 h after reperfusion.The results show that 70%hepatic ischemia-reperfusion combined with partial hepatectomy injury can lead to oxidative stress and inflammation,and its mechanism may be related to the down-regulation of antioxidant enzymes and the increased expression of pro-inflammatory factors.
