摘要
血液系统恶性肿瘤是人类常见肿瘤疾病,发病率呈逐年上升趋势。钙/钙调蛋白依赖性蛋白激酶(calcium/calmodulin-dependent protein kinase,CaMK)是广泛表达的多功能丝氨酸/苏氨酸蛋白激酶,可调节不同类型细胞的发育及生理功能。造血细胞中CaMKⅡ的主要亚型CaMKⅡγ与多种血液系统肿瘤密切相关,磷酸化CaMKⅡγ存在于多数血液肿瘤细胞中,可通过多种信号传导途径促进多种白血病干细胞增殖,抑制其分化,是调节白血病细胞增殖和分化的关键酶。研究CaMKⅡγ在血液系统肿瘤发生、发展中的分子调控机制,有望为探索血液系统肿瘤的新疗法提供一个分子靶点。本文就CaMKⅡγ在血液系统肿瘤中作用的研究进展作一综述。
Hematologic malignancy is a common tumor disease,with an annually increasing prevalence.Calcium/calmodulin-dependent protein kinase(CaMK),a widely expressed multifunctional serine/threonine protein kinase,could regulate the development and physiological functions of many different cells.CaMKⅡγ,the main subtype of CaMKⅡin hematopoietic cell,is closely correlated with a variety of hematologic malignancies.The phosphorylated CaMKⅡγexists in most tumor cells in blood,and is the key enzyme of leukemia cell proliferation and differentiation through a variety of signal transduction pathways to promote a variety of leukemia stem cell proliferation and suppress its differentiation.So an in-depth study on the molecular regulatory mechanism of CaMKⅡγon the occurrence and development of hematologic malignancies is expected to provide a molecular target for the exploration of new therapies on hematologic malignancies.This paper reviews the research progress of the role of CaMKⅡγin hematologic malignancies.
