摘要
目的探讨现代光学技术判断动物活体肾脏缺血再灌注损伤的可行性和意义及光学相干断层成像(optical coherencetomography,OCT)实现肾脏“光学活检”的能力。方法采用新型、高速、手持式的OCT系统,采集活体动物肾脏微结构的二维OCT图像及缺血再灌注前后肾脏微结构。采用MATLAB软件,利用数字图像处理中的分割算法、骨骼化等分析肾脏的结构和功能参数。结果OCT能直观观测肾脏的超微结构,包括肾小球、肾小管及血管等,OCT图像可揭示形态学相关信息。缺血前后及再灌注后相同采集点的图像区域肾小管展开数量和腔直径大小有显著变化。结论OCT成像术能观测肾脏的超微结构,可反映肾脏缺血再灌注损伤,检测移植肾活性有一定的可行性,但如何量化仍需进一步研究。
Objective To explore the feasibility and significance of modern optical technology in determining renal ischemia-reperfusion injury in living animals,and the ability of optical coherence tomography(OCT)to realizing renal"optical biopsy".Methods The 2-dimensional OCT images of renal microstructure in living animals and the changes in renal microstructure before and after ischemia-reperfusion were collected using a novel,high-speed and hand-held OCT system.The structural and functional parameters of the kidney were analyzed by the segmentation and skeletonization algorithm in digital image processing mainly using MATLAB.Results The ultrastructure of the kidney could be observed directly using OCT,including the renal glomerulus,renal tubules and blood vessels.The resolution of OCT images was enough to reveal morphological details.The number of expanded renal tubules and the diameter of renal tubules in the images with the same collecting point before and after ischemia and after reperfusion showed significant changes.Conclusion OCT imaging can display the ultrastructure of the kidney,which can reflect renal ischemia-reperfusion injury.It is feasible to detect the activity of transplanted kidney,but the quantification needs further study.
