核仁纺锤体相关蛋白1在子宫内膜癌组织中的表达及其临床意义

Expression of nucleolar spindle associated protein 1 in endometrial carcinoma and its clinical significance

目的探讨核仁纺锤体相关蛋白1(NUSAP1)在子宫内膜癌组织中的表达及与临床病理之间的关系。方法通过TCGA数据库,分析NUSAP1 mRNA在子宫内膜癌组织与正常子宫内膜组织中、配对的癌和癌旁组织中的表达情况;通过GEPIA数据库,进一步分析NUSAP1 mRNA在子宫内膜癌与正常子宫内膜组织中的表达情况;免疫组织化学法检测79例子宫内膜癌及42例正常子宫内膜组织中NUSAP1蛋白的表达水平,分析NUSAP1表达高低与子宫内膜癌患者临床病理特征之间的关系。结果TCGA数据库显示,NUSAP1 mRNA在子宫内膜癌组织中的表达明显高于正常子宫内膜组织,在配对的子宫内膜癌组织中明显高于癌旁组织(P<0.05);GEPIA数据库同样显示,NUSAP1 mRNA在子宫内膜癌组织中的表达明显高于正常子宫内膜组织(P<0.05);免疫组织化学结果显示,NUSAP1在子宫内膜癌组织中的高表达率为65.8%(52/79),而在正常子宫内膜组织中的高表达率为11.9%(5/42),差异有统计学意义(P<0.001);子宫内膜癌组织中NUSAP1蛋白高表达与组织学分级、FIGO分期和淋巴结转移有关(P<0.05)。结论NUSAP1在子宫内膜癌组织中表达升高,且其表达量高低与子宫内膜癌的进展相关,可能成为子宫内膜癌治疗的潜在靶点。

Objective To investigate the expression of nucleolar spindle associated protein 1(NUSAP1)in endometrial carcinoma(EC)tissue and its clinical significance.Methods The expressions of NUSAP1 mRNA in EC compared with normal endometrial tissues and EC compared with adjacent normal tissue were analyzed using the TCGA database.The expression of NUSAP1 in EC and normal endometrial tissues was further deter-mined via the GEPIA database.The expression of NUSAP1 protein in 79 specimens of EC and 42 normal endometrial tissues were asessed by immunohistochemistry.The correlation of NUSAP1 protein expression with clinicopathologic parameters of the patients was also analyzed.Results The TCGA database analysis revealed that the levels of NUSAP1 mRNA was highly expressed in EC compared with normal endometrial tissues and EC compared with adjacent normal tissue(P<0.05).The GEPIA database also showed that the expression of NUSAP1 mRNA in EC was significantly higher than that in normal endometrial tissues(P<0.05).The results of immunohistochemical staining showed that the rate of high NUSAP1 expression was 65.8%(52/79)in endometrial carcinoma and the rate of high NUSAP1 expression was 11.9%(5/42)in non cancerous tissues,with a significant difference between the two groups(P<0.05).Moreover,the NUSAP1 protein expression was significantly associated with the histological grade(P<0.05),FIGO stage(P<0.05),and lymph node metastasis(P<0.05).Conclusion NUSAP1 is highly expressed in endometrial carcinoma and it may be closely related to the progression of endometrial carcinoma.NUSAP1 may become an indicator of treatment target for endometrial carcinoma.
