摘要
目的分析基因HES7的表达与结直肠癌患者临床病理特征以及预后的相关性。方法从肿瘤基因组计划(TCGA)数据库下载结直肠癌组织和正常组织基因表达数据和相关的临床病理数据。利用R软件提取HES7基因表达数据,然后分析HES7在结直肠癌及癌旁组织中的差异表达。将基因HES7的表达数据分别与年龄、性别、Stage分期、TNM分期等临床病理数据合并并进行相关性分析,进一步进行多因素Cox分析和生存分析。并用GSEA富集分析软件对基因HES7进行通路富集(KEGG)分析。结果HES7在结直肠癌组织中明显高表达(W=6449.00,P<0.05);HES7低表达组中肿瘤T、N分期患者数量明显高于HES7高表达组,差异均有统计学意义(χ2分别=8.40、3.90,P均<0.05),并且HES7表达与患者性别、年龄、M分期和Stage分期无关(χ2分别=0.09、1.87、4.75、6.56,P均>0.05),基因HES7、年龄和T分期可作为评价患者预后的危险因子(HR分别=1.47、1.05、1.76,P均<0.05);HES7高表达可缩短结直肠癌患者的生存时间(χ2=5.34,P<0.05);HES7主要在过氧化物酶体、丙酸代谢、JAK/STAT信号通路、TOLL样受体信号通路等信号通路中被富集(P均<0.05),这些信号通路异常与多种恶性肿瘤的发生发展有关。结论基因HES7可能参与结直肠癌发生发展,可作为调节结直肠癌进展并影响患者预后的潜在靶标。
Objective To analyze the correlation between HES7 gene expression and clinicopathological features and prognosis of colorectal cancer patients.Methods Expression data of colorectal cancer tissues and normal tissues and rel-evant clinicopathological data were downloaded from TCGA database.R software was utilized to extract the expression data of HES7,and then to analyze the differential expression between colorectal cancer tissues and adjacent tissues.The data of HES7 gene expression was combined with clinicopathological data such as age,gender,stage and TNM stage,and the cor-relation analysis was performed.Multivariate Cox analysis and survival analysis were further performed.The KEGG of HES7 gene was analyzed by GSEA enrichment analysis software.Results HES7 was highly expressed in colorectal cancer tissues(W=6449.00,P<0.05),the patients number of HES7 in the low expression group in T and N stage were significantly higher than those of the high expression group(χ2=8.40,3.90,P<0.05),and the expression of HES7 was independent of gender,age,M stage and Stage(χ2=0.09,1.87,4.75,6.56,P>0.05).HES7 gene,age and T stage can be taken as risk factors to evaluate prognosis of colorectal cancer(HR=1.47,1.05,1.76,P<0.05),the high expression of HES7 shortened the survival time of colorectal cancer patients of colorectal cancer(χ2=5.34,P<0.05),HES7 was mainly enriched on peroxisome,propanoate metabolism,JAK/STAT signaling pathway,TOLL like receptor signaling pathway,and so on(P<0.05),whose abnormalities were related to the occurrence and development of a variety of malignant tumors.Conclusion HES7 may be engaged in the development of colorectal cancer,and may serve as a potential target for mod-ulating the progression of colorectal cancer and affecting the prognosis of patients.
