摘要
目的建立能同时测定新疆鼠尾草根中丹参素、原儿茶醛、咖啡酸及迷迭香酸4种成分质量分数的高效液相色谱法(HPLC),研究大孔吸附树脂法纯化新疆鼠尾草根中酚酸类化学成分的最优工艺。方法以4种成分的质量分数为指标,通过静态吸附率和解吸附率来评价13种型号的大孔吸附树脂对酚酸类化学成分的静态吸附性能,并在单因素实验的基础上,选取上样液质量浓度、洗脱剂体积分数和洗脱流速3个因素进行Box-Behnken中心组合设计,利用响应面法优化新疆鼠尾草根中酚酸类化学成分的纯化工艺。结果HPD600型大孔树脂的静态吸附率为70%,解吸附率为20%。由响应面实验确定的最优纯化工艺参数为:上样液质量浓度为40 mg·mL^(-1);洗脱剂体积分数为40%;洗脱流速为0.67 mL·min^(-1)。在此条件下,通过3次验证实验,测得纯化物中4种成分的质量分数之和为5.92%,是纯化前的4.4倍。结论应用响应面法优化大孔吸附树脂法纯化新疆鼠尾草根中酚酸类化学成分的工艺合理,可有效提高新疆鼠尾草根中酚酸类化学成分的质量分数。
Objective To establish a high performance liquid chromatography(HPLC)method for the determination of 4 constituents,namely 3-(3′,4′-dihydroxyphenyl)lactic acid,protocatechuic aldehyde,caffeic acid and rosmarinic acid in Salvia deserta Schang.To study the optimal purification process of phenolic acid constituents from Salvia deserta Schang by macroporous adsorption resin.Methods By taking the mass fraction of the 4 constituents as the index,static adsorption rate and adsorption rate were used to evaluate 13 types of macroporous adsorption resin for the purification of phenolic acids constituents.On the basis of single factor experiment,sample liquid mass concentration,eluent volume fraction and elution velocity were screening by the Box-Behnken central composite design and response surface method to optimize the phenolic acid constituents purification process.Results The static adsorption rate and desorption rate of HPD600 macroporous resin were 70%and 20%respectively.The optimal purification parameters determined by response surface test were as follows:mass concentration of the sample 40 mg·mL^(-1),volume fraction of the eluent 40%,and elution velocity 0.67 mL·min^(-1).Under these conditions,by 3 validation experiments,the mass fraction of the purified 4 phenolic acid constituents was 5.92%,which was 4.4 times higher than that before purification.Conclusion By using response surface method to optimize purification process of phenolic acid compounds from Salvia deserta Schang with macroporous adsorption resin is reasonable,which can effectively improve the mass fraction of phenolic acid constituents extracted from Salvia deserta Schang.
