摘要
目的建立能同时测定羽裂蟹甲草花中7-羟基香豆素(HC)、7-羟基-8-甲氧基香豆素(HMC)和7β-D-葡萄糖-8-羟基香豆素(GC)含量的高效液相色谱-二极管阵列检测器(HPLC-PDA)法。方法采用Kromasil C 18色谱柱(250 mm×4.6 mm,5μm),以甲醇-四氢呋喃-10 mL·L^(-1)醋酸(27∶7∶66)为流动相;流速:0.8 mL·min^(-1);检测波长:325 nm。结果HC、HMC和GC质量浓度分别在1~120、1~120、2~120μg·mL^(-1)范围内与峰面积呈良好的线性关系,相关系数分别为0.9998、0.9998、0.9999;平均回收率分别为101.92%、98.47%、98.94%。结论所建立的方法简便、快速、准确,可用于羽裂蟹甲草花的质量控制,并为香豆素类化合物的分离分析提供参考。
Objective To establish a high performance liquid chromatography coupled with photo-diode array(HPLC-PDA)detection method for simultaneously determining the contents of 7-hydroxy-coumarin(HC),7-hydroxy-8-methoxy-coumarin(HMC)and 7β-D-glucosyl-coumarin(GC)in the flower of Cacalia tangutica(Franch.)Hand.-Mazz..Methods The separation was achieved on a Kromasil C 18 column(250 mm×4.6 mm,5μm)using methanol-tetrahydrofuran-10 mL·L^(-1)acetic acid(27∶7∶66)as the mobile phase at a flow rate of 0.8 mL·min^(-1),and 325 nm as the detection wavelength.Results HC,HMC and GC showed good linearity in the ranges of 1-120(r 1=0.9998),1-120(r 2=0.9998)and 2-120μg·mL^(-1)(r 3=0.9999),respectively.The average recoveries were 101.92%,98.47%and 98.94%respectively.Conclusion The proposed method is simple,rapid and accurate and promises to be applicable for the quality control of flowers of Cacalia tangutica(Franch.)Hand.-Mazz.,and provide evidence for the analysis of coumarin compounds.
