响应面法结合信息熵理论优化紫姜总黄酮的提取工艺 被引量:5

摘要 目的优化紫姜药材中甲氧基黄酮的提取工艺。方法采用超声提取法,在单因素考察的基础上,结合信息熵-响应面法对乙醇体积分数、超声时间、液料比进行考察,以紫姜中6种主要甲氧基黄酮类成分的含量为评价指标,根据信息熵理论,赋予各指标权重系数,计算综合评分。通过Design-Expert 10.0.7软件分析各因素间的交互作用,得出优化的提取工艺,并对该优化工艺进行验证实验。结果优化的工艺条件为:乙醇体积分数为60%,液料比为12∶1,超声提取2次,每次25 min,甲氧基黄酮类成分的综合评分为1.024,验证实验结果为0.994,各指标成分含量及综合评分RSD值均小于1%。结论优化的提取工艺稳定可行,可为紫姜的进一步研究提供参考。 Objective To optimize the extraction process of methoxylflavones in the rhizome of Kaempferia parviflora.Methods Ultrasonication was used for the extraction.Based on single factor experiment and information entropy-response surface method,three factors including ethanol volume fraction,extraction time and liquid-to-material ratio were investigated.Taking the yields of 6 main methoxylflavones as evaluation indexes,the weight coefficient of each index was calculated according to the information entropy theory,and the comprehensive score was obtained.With Design-Expert 10.0.7 software,the interactions between the 3 factors were analyzed.The optimized extraction condition was obtained and the validation test was performed.Results The optimized extraction condition was as follows:ethanol volume fraction of 60%,liquid to material ratio of 12∶1 and extracted twice,25 minutes each times.The predicted comprehensive score was 1.024 and the verification test result was 0.994 with RSD value was less than 1%.Conclusion The optimized extraction process is stable and feasible,which can provide scientific reference for further research and development of the the rhizome of Kaempferia parviflora.
