丙酸睾酮不同给药方式对小鼠前列腺增生模型的影响研究

摘要 目的探讨采用丙酸睾酮建立小鼠前列腺增生模型的最佳给药方式。方法选取24只雄性ICR小鼠,随机分为腹腔注射空白组、腹腔注射模型组、皮下注射空白组、皮下注射模型组,每组6只。腹腔注射模型组和皮下注射模型组按照相应方法注射丙酸睾酮12.5 mg/(kg·d),腹腔注射空白组和皮下注射空白组按照相应方法注射等量生理盐水,均连续注射31 d。末次给药完毕禁食不禁水12 h,用摘眼球取血法取血检测睾酮(T)和雌二醇(E_(2))水平,计算T/E_(2);脱颈椎处死小鼠,解剖分离前列腺、胸腺及脾脏,计算各脏器指数,并在光镜下观察前列腺组织病理学特点。结果腹腔注射模型组小鼠E_(2)水平显著高于腹腔注射空白组(P<0.05),T水平与T/E_(2)与腹腔注射空白组比较差异均无统计学意义(P均>0.05);皮下注射模型组小鼠T、E_(2)水平及T/E_(2)均显著高于皮下注射空白组(P均<0.05)。腹腔注射模型组小鼠前列腺湿重及前列腺指数均显著高于腹腔注射空白组(P均<0.05),皮下注射模型组均显著高于皮下注射空白组(P均<0.05);腹腔注射模型组小鼠胸腺与脾脏指数与腹腔注射空白组比较差异均无统计学意义(P均>0.05);皮下注射模型组小鼠胸腺指数显著低于皮下注射空白组(P<0.05),脾脏指数与皮下注射空白组比较差异无统计学意义(P>0.05)。腹腔注射模型组小鼠前列腺个别腺腔扩张,间质中有少量纤维结缔组织及平滑肌增生,偶见血管充血;皮下注射模型组小鼠前列腺腺腔明显扩张,间质内纤维结缔组织及平滑肌明显增生,血管充血明显。结论在保证其他造模条件不变的情况下,腹腔注射与皮下注射丙酸睾酮均可建立小鼠前列腺增生模型,但皮下注射建立的前列腺增生模型特征更为显著。 Objective It is to explore the optimal administration method for the establishment of mouse model of prostatic hyperplasia(BPH)with testosterone propionate.Methods Twenty-four male ICR mice were selected and randomly divided into an intraperitoneal injection blank group,an intraperitoneal injection model group,a subcutaneous injection blank group,and a subcutaneous injection model group,with 6 mice in each group.The intraperitoneal injection model group and the subcutaneous injection model group were injected with testosterone propionate 12.5 mg/(kg·d)according to the corresponding method.The intraperitoneal injection blank group and the subcutaneous injection blank group were injected with the same amount of normal saline according to the corresponding method,and they were injected continuously for 31 days.After the last administration,fasting without water for 12 hours,the blood was taken from the eyeball to detect the levels of testosterone(T)and estradiol(E_(2)),and to calculate T/E_(2);the mice were sacrificed by cervical vertebrae,and their prostate,thymus and spleen were dissected and separated.The index of each organ was calculated,and the histopathological characteristics of the prostate was observed under a light microscope.Results The E_(2) level of mice in the intraperitoneal injection model group was significantly higher than that in the intraperitoneal injection blank group(P<0.05),and there was no significant difference in T level and T/E_(2) between the two groups(P>0.05);the levels of T,E_(2) and T/E_(2) of mice in the subcutaneous injection model group were significantly higher than those in the subcutaneous injection blank group(all P<0.05).The wet prostate weight and prostate index of the mice in the intraperitoneal injection model group were significantly higher than those in the intraperitoneal injection blank group(all P<0.05),and the subcutaneous injection model group was significantly higher than the subcutaneous injection blank group(all P<0.05);there was no significant difference in mouse thymus and spleen index between the intraperitoneal injection model group and intraperitoneal injection blank group(P>0.05);the thymus index in the subcutaneous injection model group mouse was significantly lower than that in the subcutaneous injection blank group(P<0.05),but there was no significant difference in the spleen index between the two groups(P>0.05).In the intraperitoneal injection model group,individual glandular cavities of the prostate were dilated,and there was a small amount of fibrous connective tissue and smooth muscle hyperplasia in the interstitium,and vascular congestion was occasionally found;in the subcutaneous injection model group,the prostate gland cavities of the mice were significantly expanded,and the interstitial fibrous connective tissue and smooth muscle were obvious hyperplasia,vascular congestion was obvious.Conclusion Under the condition that other modeling conditions remain unchanged,both intraperitoneal injection and subcutaneous injection of testosterone propionate can establish a mouse BPH model,but the characteristics of the BPH model established by subcutaneous injection are more significant.
