黄芩汤对湿热型溃疡性结肠炎大鼠结肠组织中NF-κB p65和ICAM-1表达的影响 被引量:7

摘要 目的观察黄芩汤对湿热型溃疡性结肠炎(UC)模型大鼠结肠组织中NF-κB p65和ICAM-1表达的影响,探讨黄芩汤治疗UC的作用机理。方法将80只SD大鼠随机分为正常对照组、模型组、黄芩汤组、柳氮磺吡啶组,除对照组外,采用高脂高糖辛辣饮食加2,4,6三硝基苯磺酸(TNBS)诱导法构建湿热型UC大鼠模型,造模成功后,分别给予20 mL·kg^(-1)体质量的黄芩汤和8%浓度的柳氮磺砒啶混悬液灌胃治疗,正常对照组和模型组给予等量的生理盐水,连续治疗2周,实时荧光定量聚合酶链式反应检测结肠组织中NF-κB p65和ICAM-1 mRNA表达。免疫组化检测结肠组织中NF-κB p65和ICAM-1蛋白表达。结果与正常对照组比较,模型组NF-κB p65和ICAM-1 mRNA及蛋白表达量明显升高(P<0.05);与模型组比较,黄芩汤组和柳氮磺砒啶组NF-κB p65和ICAM-1 mRNA及蛋白表达量显著下降(P<0.05);黄芩汤组NF-κB p65和ICAM-1 mRNA表达量及蛋白阳性表达率显著低于柳氮磺砒啶组,2组比较差异具有统计学意义(P<0.05)。结论黄芩汤对湿热型溃疡性结肠炎的治疗作用可能是通过下调NF-κB p65、ICAM-1的表达来实现。 Objective:To observe the effect of Scutellaria baicalansis Decoction on the expression of NF-κB p65 and ICAM-1 in the colon tissue of a rat model of damp-heat ulcerative colitis(UC),and to explore the mechanism of Scutellaria Decoction in treating UC.Methods:80 SD rats were randomly divided into normal control group,model group,Scutellaria Decoction group and sulfasalazine group.Except for the control group,a high-fat,high-sugar spicy diet plus 2,4,6 trinitro-benzene sulfonic acid(TNBS)were used to construct a damp-heat UC rat model.After the model was successfully established,the rats were given by intragastric administration of 20 mLokg-1 body weight Scutellaria Decoction and 8% concentration of sulfasalazine suspension.The normal control group and the model group were given the same amount of normal saline for 2 successive weeks and the real-time fluorescence quantitative polymerase chain reaction was used to detect the expression of NF-κB p65 and ICAM-1 mRNA in the colon tissue and the immunity class chemically detected the NF-κB p65 and ICAM-1 protein expression in the colon tissue.Results:Compared with that of the normal control group,the NF-κB p65 and ICAM-1 mRNA and protein expression quantity of the model group was significantly increased(P<0.05);compared with that of the model group,the NF-κB p65and ICAM-1 mRNA and protein quantity expression of the Scutellaria Decoction group and the sulfasalazine group decreased significantly(P<0.05),The NF-κB p65 and ICAM-1 mRNA expression quantity and protein positive expression rate of the Scutellaria Decoction group was significantly lower than that of the sulfasalazine group.The difference was statistically significant between the two groups(P<0.05).Conclusion:The curative effect of Scutellaria Decoction on damp-heat ulcerative colitis may be achieved by down-regulating the expression of NF-κB p65 and ICAM-1.
