Optimization of Extraction Process of Concentrated Decoction of Qingqi Jiedu Mixture by Multi-Index Orthogonal Test

目的优选清气解毒合剂浓煎剂的提取工艺。方法以加水量、煎煮时间、煎煮次数为考察因素,以干膏得率及柴胡皂苷a、连翘苷、连翘酯苷A含量为评价指标,采用L9(34)正交试验法优选清气解毒合剂浓煎剂的提取工艺。结果最佳提取工艺为加12倍量水每次煎煮2 h,煎煮3次。干膏得率大于26%,柴胡皂苷a、连翘苷、连翘酯苷A含量分别大于1.9,3.8,2.1 mg/g。结论该优选的工艺适合清气解毒合剂浓煎剂制备的生产,可操作性强,为未来的大生产提供了参考。

Objective To optimize the extraction process of concentrated decoction of Qingqi Jiedu Mixture.Methods The extraction process of concentrated decoction of Qingqi Jiedu Mixture was optimized by L9(34)orthogonal test with water addition,decocting time and decocting times as the investigation factors,with dry extract yield and the content of saikosaponin a,forsythin and forsythiaside A as the evaluation indexes.Results The best extraction process was decoction for 3 times with 2 h each time and adding 12 times the amount of water.The dry extract yield was higher than 26%,and the contents of saikosaponin a,forsythin and forsythiaside A were more than 1.9,3.8 and 2.1 mg/g,respectively.Conclusion The optimized process is suitable for the preparation of concentrated decoction of Qingqi Jiedu Mixture,which has strong operability and provides a reference for large-scale production in the future.
